部分细菌和真菌特征性挥发性有机物的研究

【摘要】:目的: 通过检测部分常见的细菌与真菌菌株代谢产生的挥发性有机物(volatile organic compounds VOCs),筛选出各菌株产生的内源性VOCs,为呼吸气体在肺部感染性疾病的诊断提供实验依据。 方法: 利用固相微萃取联合气相色谱质谱仪连用技术(solid-phase microextraction and gas chromatography mass spectroscopy,SPME/GC-MS),检测体外培养菌株混悬液(大肠杆菌、铜绿假单胞菌、肺炎克雷伯杆菌、金黄色葡萄球菌、肠球菌、白色念珠菌、光滑念珠菌)顶空气体中的VOCs,从中筛选出各种病原菌的特征性VOCs。 结果: 试验中大肠杆菌、铜绿假单胞菌、肺炎克雷伯杆菌及白假丝酵母菌可检测到特征性VOCs,大肠杆菌、铜绿假单胞菌及肺炎克雷伯杆菌中可分别检测到特征性吲哚、十一烷醇及丁基羟基甲苯,白假丝酵母菌可检测到C15H260和C17H2802;大肠杆菌与铜绿假单胞菌的混合菌悬液可检测到吲哚及十一烷醇,与各自培养检测到的VOC相同,相互之间无影响;细胞株与微生物混合培养,结果与单纯微生物培养的VOCs相同,提示细胞株不影响微生物代谢;白假丝酵母菌与光滑念珠菌的特征性VOCs不同,提示通过检测细菌与真菌特征性VOCs不仅能鉴别菌属,且能鉴别相近的菌种。 结论: 部分体外培养的细菌与真菌可通过代谢产生特征性VOCs,两种细菌混合生长仍可产生各自的特征性VOCs,A549细胞株不影响细菌特征性VOCs;SPEM/GC-MS是一种可用于检测部分细菌和真菌特征性VOCs的无创快速有效的方法,且可鉴别相近的菌种。
[Abstract]:Objective:. By detecting volatile organic compounds VOCs produced by metabolism of some common bacteria and fungal strains, the endogenous VOCs produced by each strain were screened, which provided experimental basis for the diagnosis of respiratory gas in pulmonary infectious diseases. Methods:. Solid phase microextraction and gas chromatography mass spectroscopy SPME / GC MS / MS was used to detect the suspension (E. coli, Pseudomonas aeruginosa, Klebsiella pneumoniae, Staphylococcus aureus, Enterococcus) in vitro by solid phase microextraction (SPME) combined with gas chromatographic mass spectrometry (GC). The characteristic VOCs in the headspace gas of Candida albicans were screened out. Results:. E. coli, Pseudomonas aeruginosa, Klebsiella pneumoniae and Candida albicans could be detected in E. coli, E. coli, Pseudomonas aeruginosa and Klebsiella pneumoniae respectively. C15H260 and C17H2802 could be detected in undecanol and Ding Ji hydroxy toluene, C15H260 and C17H2802 could be detected in Escherichia coli and Pseudomonas aeruginosa suspension, indole and undecanol could be detected in mixed suspension of Escherichia coli and Pseudomonas aeruginosa, which were the same as VOC detected in culture and had no effect on each other. The results of mixed culture of cell line and microorganism were the same as that of VOCs cultured by pure microorganism, suggesting that the cell strain had no effect on microbial metabolism, and the characteristic VOCs of Candida albicans and Candida glabra were different. It is suggested that the characteristic VOCs of bacteria and fungi can not only identify the genus of bacteria, but also identify the similar species. Conclusion:. Some of the bacteria and fungi cultured in vitro can produce characteristic VOCs by metabolism, and the mixed growth of the two bacteria can still produce their own characteristic VOCsSU A549 cell lines, which can be used for the detection of some bacteria and fungi. A noninvasive, fast and effective method for characteristic VOCs, And the similar strains can be identified.
