三味干姜散对两种实验性肝损动物模型的调控作用研究

【摘要】:目的从保护肝脏线粒体和调控"Nrf2/Bach1-ARE-抗氧化酶"通路两个角度,观察三味干姜散对实验性肝损伤动物模型的影响。方法复制大鼠急性酒精性肝损伤模型和小鼠刀豆蛋白A(Con A)免疫性肝损伤模型,分别给予三味干姜散后,检测肝功能指标ALT、AST,进行病理学观察;提取肝线粒体,检测线粒体膜电位、MPTP、SOD、8-OHd G、呼吸链复合物Ⅰ、Ⅲ酶活性以及肝组织中MDA、GSH、GSH-Px水平;采用RT-PCR检测肝组织中Nrf2、Bach1、HO-1的m RNA表达水平。结果对于急性酒精性肝损伤模型,三味干姜散能降低ALT、AST(P0.05)和8-OHd G(P0.01)以及MDA的含量(P0.05,P0.01),提高肝组织中抗氧化剂GSH、SOD和GSH-Px酶的活性(P0.05,P0.01),上调Nrf2和HO-1 m RNA的表达(P0.05),显著增加呼吸链复合物Ⅰ、Ⅲ酶的活性(P0.01),以及降低线粒体膜通道孔的通透性(P0.05);对于免疫性肝损伤模型,三味干姜散能降低ALT、AST水平和MDA活性(P0.05,P0.01),升高SOD(P0.05)、GSH-Px与GSH酶的活性(P0.05,P0.01),显著上调线粒体膜电位(P0.01)。结论三味干姜散对大鼠急性酒精性肝损伤和小鼠Con A免疫性肝损伤均有一定的保护作用,其机制与三味干姜散抗肝脏线粒体损伤和调控"Nrf2/Bach1-ARE-抗氧化酶"通路有关。
[Abstract]:Objective to observe the effect of Sanwei Ganjiang Powder on experimental liver injury from two aspects: protecting liver mitochondria and regulating "Nrf2 / Bach1-ARE- antioxidant enzyme" pathway.Methods the acute alcoholic liver injury model in rats and the immunological liver injury model in mice with concanavalin A(Con A were established. The liver function indexes of alt AST were detected and the liver mitochondria were extracted after three kinds of dry ginger powder were given respectively.Conclusion Sanwei Ganjiang Powder has protective effects on acute alcoholic liver injury in rats and Con A immune liver injury in mice, and its mechanism is related to the regulation of "Nrf2 / Bach1-ARE- antioxidant enzyme" pathway by Sanwei Ganjiang Powder in preventing liver mitochondria damage and in regulating "Nrf2 / Bach1-ARE- antioxidant enzyme" pathway.
