家兔半膜肌各亚部肌球蛋白重链表达差异的实验研究

【摘要】:目的骨骼肌肌纤维类型的构成比较复杂,呈单一性分布的较少。人们发现家兔半膜肌各个亚部的肌纤维构成较其它骨骼肌单一。本实验在此基础上通过肌球蛋白重链的亚型的分类方法检测家兔半膜肌各亚部的肌纤维型的构成情况,从分子水平观察其肌纤维类型的分布特点,为进一步研究两型肌纤维在结构、能量代谢、基因表达和功能等方面的区别以及两型肌纤维转化机制奠定基础。 方法实验以10只成年家兔为研究对象,雌雄不拘,取其右侧肢体半膜肌。通过改良肌球蛋白ATP酶染色的方法验证其四个亚部两型肌纤维分布情况及构成比例,同时采用反转录聚合酶链式反应(RT-PCR)检测各个亚部MHCⅠ、MHCⅡa、MHCⅡb、及MHCⅡx四个亚型mRNA的构成及含量。 结果家兔半膜肌四个亚部中,固有半膜肌只含有慢肌(Ⅰ型肌纤维),表达MHCⅠ型和少量MHCⅡa型基因。副半膜肌腹侧含有少量慢肌(Ⅰ型肌纤维),表达少量MHCⅠ型基因,其余主要含有快肌(Ⅱ型肌纤维),表达MHCⅡa、MHCⅡb和MHCⅡx型基因;而其背侧和外侧只含有快肌,仅表达MHCⅡa、MHCⅡb和MHCⅡx型基因。 结论固有半膜肌亚部为慢肌,含有慢肌纤维,即Ⅰ型肌纤维,主要表达MHCⅠ和MHCⅡa两个亚型。副半膜肌的三个亚部中,除腹侧亚部表达极少量MHCⅠ亚型,均主要表达MHCⅡa、MHCⅡb和MHCⅡx亚型。背侧和外侧亚部不含有慢肌纤维(Ⅰ型肌纤维),不表达MHCⅠ亚型。
