氯胺酮完全抗原的制备及免疫效果检测

【摘要】:基于盐酸氯胺酮是一种化学反应性质活泼的小分子,利用半抗原—载体效应的免疫学原理,以三氯化磷(PCl3)为桥梁,将盐酸氯胺酮分子共价连接到兔血清白蛋白(RSA)分子,随后采用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)分析偶联物,并用该偶联物免疫BALB/c小鼠,最后采用斑点酶联免疫吸附试验(dot-ELISA)检测氯胺酮抗体应答水平。蛋白电泳分析显示已成功制备出分子量约为78.8 ku、氯胺酮与RSA的分子偶联比为39的完全抗原,能诱导实验小鼠产生效价为1∶1 280的抗氯胺酮抗体。本试验不仅建立了检测氯胺酮的血清学方法,还为开发氯胺酮戒毒疫苗做了有益的积累。
[Abstract]:Based on the fact that ketamine hydrochloride is a small molecule with active chemical reaction properties, using the immunological principle of hapten carrier effect, the ketamine hydrochloride molecule is covalently linked to rabbit serum albumin (RSA) molecule by using phosphorous trichloride (PCL _ 3) as a bridge.BALB/c mice were immunized with sodium dodecyl sulfate and polyacrylamide gel electrophoresis (SDS-PAGE). Finally, dot-ELISAs were used to detect the level of ketamine antibody response.Electrophoretic analysis showed that the complete antigen with molecular weight of 78.8 and molecular coupling ratio of Ketamine to RSA of 39 was successfully prepared, which could induce the production of anti-ketamine antibody with a titer of 1:1 280 in experimental mice.This study not only established a serological method for the detection of ketamine, but also made a beneficial accumulation for the development of ketamine abstinence vaccine.
