大豆异黄酮对子宫内膜异位症大鼠模型中Foxp3及Cox-2表达的影响

【摘要】:目的:探讨大豆异黄酮(Soybean Isoflavone, SIF)对子宫内膜异位症(endometriosis, EMs)大鼠模型中Foxp3及COX-2表达的影响。 方法:将50只Wistar雌性大鼠采用外科手术大鼠自体子宫内膜移植法建立大鼠子宫内膜异位症模型,随机分对照组、大豆异黄酮高剂量组、大豆异黄酮低剂量组。造模成功后给药6周,并做病理学检查。观察比较治疗前后移植物体积大小,并通过免疫组织化学SABC法检测大鼠异位内膜组织中Foxp3及COX-2的表达。 结果:1.移植物外观:对照组与SIF低剂量组移植物体积无明显差异(P0.05),SIF高剂量组移植物体积明显缩小(PO.01); 2.病理学改变:与对照组比较,治疗组移植腺体上皮细胞变小上皮细胞层厚度变薄,其中SIF高剂量组效果比SIF低剂量组明显; 3.与对照组比较,治疗组腺体中Foxp3表达率明显降低(P0.05),且SIF高剂量组中Foxp3的下降程度比SIF低剂量组的更明显(P0.05); 4.与对照组比较,治疗组腺体中COX-2表达率明显降低(P0.01);且SIF高剂量组COX-2表达的下降程度比SIF低剂量组更明显(P0.05); 5.异位症内膜中Foxp3的表达与COX-2的表达呈正相关(r=0.881,P0.01)。 结论:1.高剂量的大豆异黄酮对大鼠子宫内膜异位症模型中的异位内膜组织具有明显的抑制作用,其机制可能与大豆异黄酮下调了异位组织中COX-2及Foxp3的表达,从而抑制异位组织中新生血管的生成及提高机体免疫监视功能有关。 2.大鼠EMs的异位内膜中COX-2的表达与Foxp3的表达呈正相关,提示血管生成及免疫抑制因素在EMs的发生机制中起协同作用。
[Abstract]:Aim: to investigate the effect of soybean isoflavone Isoflavone (siff) on the expression of Foxp3 and COX-2 in endometriosis rat model.Methods: 50 Wistar female rats were randomly divided into control group, high dose daidzein group and low dose daidzein group.The model was successfully treated for 6 weeks, and pathological examination was made.The volume of grafts before and after treatment was observed and the expression of Foxp3 and COX-2 in ectopic endometrium of rats was detected by immunohistochemical SABC method.The result is 1: 1.Graft appearance: there was no significant difference in graft volume between the control group and the low dose SIF group.2.Pathological changes: compared with the control group, the thickness of epithelial cell layer of transplanted glandular epithelial cells in the treatment group was thinner, and the effect of SIF in high dose group was more obvious than that in SIF low dose group.3.Compared with the control group, the expression rate of Foxp3 in the glands of the treatment group was significantly lower than that of the control group, and the decrease of Foxp3 in the high dose group of SIF was more significant than that in the low dose group of SIF.4.Compared with the control group, the expression rate of COX-2 in the glands of the treatment group was significantly lower than that of the control group, and the decrease of COX-2 expression in the high dose group of SIF was more significant than that in the low dose group of SIF.5.There was a positive correlation between the expression of Foxp3 and COX-2 in endometrium.Conclusion 1.High dose soybean isoflavones could inhibit the expression of COX-2 and Foxp3 in the ectopic endometrium of rats, and the mechanism might be related to the down-regulation of the expression of COX-2 and Foxp3 in the ectopic tissues by soybean isoflavones.It is related to the inhibition of angiogenesis in ectopic tissues and the enhancement of immune surveillance function.2.The expression of COX-2 in ectopic endometrium of rat EMs was positively correlated with the expression of Foxp3, suggesting that angiogenesis and immunosuppressive factors play a synergistic role in the pathogenesis of EMs.
