吞噬及饥饿诱导巨噬细胞自噬对其吞噬功能的影响

【摘要】:目的研究饥饿诱导的自噬对巨噬细胞吞噬鸡红细胞的影响,初步探讨自噬在巨噬细胞吞噬功能中的作用。方法实时动态观察巨噬细胞吞噬鸡红细胞的过程,单丹磺酰尸胺(MDC)荧光染色标记巨噬细胞的自噬体,激光扫描共聚焦显微镜下进行观察和定量分析。Western blot方法检测巨噬细胞的微管相关蛋白1轻链3(LC3)的表达,HE染色观察自噬对其吞噬功能的影响。结果巨噬细胞活跃地吞噬鸡红细胞,吞噬后6 h MDC荧光染色阳性率明显高于对照组。饥饿诱导巨噬细胞LC3Ⅱ表达增高,并可以被3-甲基腺嘌呤(3-MA)抑制,饥饿3 h后的巨噬细胞对鸡红细胞的吞噬率与对照组相比无明显变化。自噬被3-MA抑制后,巨噬细胞对鸡红细胞的吞噬率明显低于对照组。结论巨噬细胞活跃地吞噬鸡红细胞,吞噬后自噬明显增强;自噬对巨噬细胞吞噬功能有重要作用。
[Abstract]:Objective to study the effect of starvation induced autophagy on phagocytosis of chicken erythrocytes and the role of autophagy in phagocytosis of macrophages.Methods the process of phagocytosis of chicken erythrocytes by macrophages was observed in real time.The effect of autophagy on phagocytosis was observed by HE staining and the expression of microtubule-associated protein 1 light chain 3LC-3 in macrophages was detected by Western blot.Results the positive rate of MDC fluorescence staining of macrophages was significantly higher than that of the control group 6 h after phagocytosis.The expression of LC3 鈪,
