人精液凝固蛋白I衍生合成肽Sg I-52与精子活动关系的初步研究

【摘要】:目的:研究人精液凝固蛋白Ⅰ小分子衍生合成肽SgⅠ-52对精子活动的影响并初步探讨其可能的机制。 方法:用Percoll不连续密度梯度离心法筛选出正常精液中的活动精子,在不去膜以及去膜的处理后,与不同浓度的SgⅠ-52进行不同时间的作用,在计算机辅助精液分析(CASA)下观察精子活动的变化,用SPSS软件进行统计分析,所得结果结合透射电子显微镜下未去膜精子及去膜精子结构的特点,对SgⅠ小分子衍生合成肽SgⅠ-52对精子活动产生影响的机制进行初步探讨。 结果:去膜精子试验中,SgⅠ-52的100pg/个精子和200pg/个精子两种浓度在90min内都使精子活动的各项参数明显下降(P<0.05);不同浓度SgⅠ-52对去膜精子活动影响的差异表现为VCL和BCF参数的差异(P<0.05);在各个浓度组内,数据的变化与作用时间明显相关。 未去膜精子试验中,100pg/个精子浓度的SgⅠ-52在90min内使精子的活率下降(P<0.05),200pg/个精子浓度的SgⅠ-52使活率、A级、B级以及ALH、STR下降(P<0.05);不同浓度SgⅠ-52对未去膜精子活动影响的差异表现为VCL、VSL和BCF参数的差异(P<0.05);在各个浓度组内,数据的变化亦与作用时间明显相关。 去膜精子和未去膜精子试验比较,去膜精子组精子活动参数的改变与浓度的相关性要小于未去膜精子组,表现为去膜精子试验中不同浓度间精子活动参数的变化要小,低浓度的SgⅠ-52就能明显抑制去膜精子的活动。 结论:SgⅠ-52对去膜精子和未去膜精子的活动均有明显的抑制活性,该抑制活性呈剂量/时效相关性,且SgⅠ-52对去膜精子的抑制效应明显强于对未去膜精子的。由此推测细胞膜的完整性对于精子的正常活动十分重要,SgⅠ-52可能作用于细胞膜抑制精子的活动;对于膜被破坏但内部结构完整的活动精子,SgⅠ-52可能作用于细胞内抑制精子的活动。
[Abstract]:Objective: To study the effect of human seminal coagulin protein I Sg I -52 on sperm motility and to explore its possible mechanism.
Methods: using Percoll discontinuous density gradient centrifugation were normal in semen sperm, not to go in film and film, with different concentrations of Sg I -52 for different time, analysis in computer aided semen (CASA) changes were observed under the activity of the sperm, analyzed by SPSS the results of software, combined with the characteristics of the transmission electron microscope did not go to the sperm membrane and the sperm membrane structure, mechanism of influence on Sg of small molecule derived synthetic peptides of Sg I -52 on the activity of the sperm was investigated.
Results: the sperm membrane test, Sg I -52 100pg/ 200pg/ two sperm and sperm concentration in 90min the parameters of sperm activity decreased significantly (P < 0.05); the difference between effects of different concentrations of Sg I -52 of demembranated sperm activity showed VCL and BCF parameters (P < 0.05); in each concentration group, was significantly correlated with the changes of time data.
Not to test the sperm membrane, sperm concentration 100pg/ Sg 1 -52 in 90min in the live rate of sperm decreased (P < 0.05), 200pg/ Sg I -52 the sperm concentration, viability, Grade A, B and ALH, STR decreased (P < 0.05); different concentrations of Sg on -52 1 go to the sperm membrane induced differences of VCL between VSL and BCF parameters (P < 0.05); in each concentration group, the change of the data was also correlated with the duration of action.
Compared to the sperm membrane and not to the sperm membrane test, correlation and concentration of demembranated sperm motility parameters of the group is smaller than the demembranated sperm group, changes in the performance of the sperm activity parameters among different concentration to the sperm membrane test in small, low concentration of Sg of -52 can inhibit the sperm membrane activity.
Conclusion: Sg I -52 on to the sperm membrane and not to the sperm membrane activity had inhibitory activity, dose / aging correlation between the inhibitory activity and the inhibitory effect of Sg on the -52 to the sperm membrane was stronger than to go to the sperm membrane integrity. Which indicated that the cell membrane is very important for the normal activities sperm, Sg I -52 may function in the cell membrane to inhibit sperm motility; membrane damage but complete structure of sperm activity, Sg I -52 may play a role in intracellular inhibition of sperm motility.
