下调CDH17基因对cisplatin耐药性人胃癌BGC823细胞裸鼠移植瘤模型的影响

【摘要】:目的研究下调CDH17基因对cisplatin耐药的人胃癌BGC823细胞裸鼠移植瘤模型的影响,从体内实验研究治疗胃癌新的治疗手段。方法以人胃癌BGC823细胞为研究材料,建立人胃癌BGC823细胞裸鼠移植瘤模型。同时,构建干扰质粒从基因和蛋白水平对CDH17进行干扰,成功构建慢病毒载体。将细胞分为三组,分别是WT对照组,si NC阴性对照组,si CDH17干扰组。通过TUNEL方法和Annexin V/PI双染色法检测各组细胞的凋亡水平。结果 Real-time PCR和蛋白免疫印迹实验证实成功构建了CDH17的干扰表达载体。TUNEL检测实验表明下调CDH17基因可以促进对cisplatin耐药的人胃癌BGC823细胞裸鼠移植瘤模型细胞凋亡。Annexin V/PI双染色检测si CDH17干扰组细胞早期凋亡率远高于WT对照组和si NC阴性对照组的细胞。结论下调CDH17基因可以促进cisplatin耐药性人胃癌BGC823细胞凋亡,抑制肿瘤细胞增殖而达到治疗胃癌的目的。
[Abstract]:Objective to study the effect of down-regulation of CDH17 gene on the transplanted tumor model of human gastric cancer BGC823 cells resistant to cisplatin in nude mice, and to study a new therapeutic method for gastric cancer in vivo. Methods Human gastric cancer BGC823 cells were used as materials to establish nude mice model of transplanted tumor of human gastric cancer BGC823 cells. At the same time, the interference plasmid was constructed to interfere with CDH17 from gene and protein levels, and the lentivirus vector was successfully constructed. The cells were divided into three groups, namely WT control group and control group. The apoptosis level was detected by TUNEL and Annexin V/PI double staining. Results Real-time PCR and Western blot confirmed that the interference expression vector of CDH17 was successfully constructed. Tunel assay showed that down-regulation of CDH17 gene could promote apoptosis. Annexin V/PI of cisplatin resistant human gastric cancer BGC823 cells in nude mice. The early apoptosis rate of si CDH17 interference group was much higher than that of WT control group and si NC negative control group. Conclusion down-regulation of CDH17 gene can promote the apoptosis of BGC823 cells and inhibit the proliferation of gastric cancer cells in cisplatin resistant human gastric cancer.
