多烯磷脂酰胆碱对树突状细胞的抑制作用研究

【摘要】:目的研究多烯磷脂酰胆碱(PPC)对树突状细胞(DC)表型分化及分泌炎症因子的影响,探讨PPC抗炎作用。方法 8只SPF纹雌性Balb/c小鼠,随机分为四组,每组2只。运用免疫磁珠分析小鼠脾脏DC,分别加入PPC(PPC组,2只)、脂多糖(LPS,LPS组,2只)、PPC+LPS(PPC+LPS组,2只)以及磷酸盐缓冲液(PBS,PBS组,2只)进行刺激。24 h后收集细胞及培养上清,流式细胞术检测DC表面活化分子表达情况,流式微珠阵列法(CBA)检测细胞因子分泌情况。结果 PPC组DC表面CD40、CD80、CD86、主要组织相容性复合体(MHC)-Ⅱ分别为(23.43±2.79)%、(19.41±1.33)%、(44.30±3.16)%、(80.87±1.32)%,低于PBS组的(29.10±8.14)%、(31.81±1.02)%、(47.70±6.32)%、(83.17±2.84)%;且显著低于LPS组的(42.33±0.58)%、(63.57±2.61)%、(56.07±0.06)%、(86.20±0.95)%(P0.01);PPC+LPS组CD40、CD80、CD86、MHC-Ⅱ分别为(16.73±3.14)%、(16.12±1.92)%、(43.60±0.95)%、(78.83±1.46)%,显著低于LPS组(P0.01)。PPC组和PPC+LPS组白细胞介素(IL)-6、肿瘤坏死因子(TNF)显著低于LPS组(P0.01)。PPC+LPS组单核细胞趋化蛋白-1(MCP-1)水平明显低于LPS组(P0.01)。结论 PPC可通过抑制DC活化及炎症因子释放,发挥抗炎效果。
[Abstract]:Objective to investigate the effect of polyenylphosphatidylcholine (PPC) on phenotype differentiation and secretion of inflammatory cytokines in dendritic cells (DC). Methods eight SPF striated female Balb/c mice were randomly divided into four groups with 2 mice in each group. The spleen of mice was analyzed by immunomagnetic beads. The cells were collected and cultured in PPC(PPC group (n = 2), lipopolysaccharide (LPS) group (n = 2) and PPC LPS(PPC LPS group (n = 2) and phosphate buffer solution (PBS) group (n = 2).
