半夏白术天麻汤对痰湿壅盛型高血压模型大鼠下丘脑蛋白质组学的影响

【摘要】:目的探讨半夏白术天麻汤治疗痰湿壅盛型高血压病的可能分子机制。方法大鼠高脂饮食24周建立痰湿壅盛型高血压模型,16只造模成功大鼠随机分为模型组和半夏白术天麻汤组,每组各8只,并以普通饲料喂养大鼠8只设为正常组。半夏白术天麻汤组给予半夏白术天麻汤13.8 g/(kg·d)灌胃,正常组和模型组给予等量生理盐水灌胃,均灌胃12周。记录大鼠体重、血压,计算Lee's指数,检测大鼠血清葡萄糖、甘油三酯、总胆固醇水平;双向电泳分离样品并且进行硝酸银染色,筛选下丘脑差异蛋白质点提交质谱鉴定,采用MALDI-TOF-TOF质谱仪进行质谱分析、肽指纹图谱进行蛋白质鉴定,选取部分蛋白质点验证蛋白质组学结果。结果半夏白术天麻汤组大鼠体重、血压、Lee's指数、血清总胆固醇均较模型组下降(P0.05)。正常组、模型组、半夏白术天麻汤组分别分离出3342、3844、3828个蛋白点,其中半夏白术天麻汤组同模型组比较,100个蛋白质表达降低,72个蛋白质表达增加。在质谱仪进一步鉴定的19种蛋白中,模型组与正常组相比有差异而半夏白术天麻汤可使差异趋势逆转的蛋白点为11个,其中5个表达上调,6个表达下调。结论半夏白术天麻汤治疗痰湿壅盛型高血压病的分子机制可能与改善细胞骨架和形态、内质网应激、能量代谢、神经元结构等有关。
[Abstract]:Objective to explore the possible molecular mechanism of Banxia Atractylodes Tianma decoction in the treatment of hypertensives with phlegm and dampness. Methods Sixteen rats were randomly divided into two groups: model group and Banxia Atractylodes Tianma decoction group, with 8 rats in each group, and 8 rats were fed with normal diet as normal group. Banxia Atractylodes Tianma decoction group was given Banxia Atractylodes Tianma decoction 13.8 g/(kg d), normal group and model group were given the same amount of normal saline for 12 weeks. Body weight, blood pressure, Lee's index, serum glucose, triglyceride and total cholesterol levels were measured, samples were separated by two-dimensional electrophoresis and silver nitrate staining was performed to screen hypothalamic differential protein spots for mass spectrometry. MALDI-TOF-TOF mass spectrometer was used for mass spectrometry, peptide fingerprint was used for protein identification, and some protein spots were selected to verify the proteomic results. Results the body weight, blood pressure and serum total cholesterol of Banxia Atractylodes Tianma decoction group were lower than that of model group (P 0.05). In normal group, model group and Banxia Atractylodes Tianma decoction group, 3342 protein spots were isolated, 3828 protein spots were isolated respectively. Compared with model group, 100 protein expression decreased and 72 protein expression increased in Banxia Atractylodes Tianma decoction group. Among the 19 proteins further identified by mass spectrometer, there were differences between the model group and the normal group, while the protein points of Banxia Atractylodes Tianma decoction could reverse the trend of the difference were 11, among which 5 were up-regulated and 6 down-regulated. Conclusion the molecular mechanism of Banxia Atractylodes Tianma decoction in the treatment of hypertensives with phlegm-dampness accumulation may be related to the improvement of cytoskeleton and morphology, endoplasmic reticulum stress, energy metabolism and neuronal structure.
