腹膜纤维化大鼠模型构建及其腹膜组织中热休克蛋白的表达变化

【摘要】:目的构建腹膜纤维化大鼠模型,探讨其腹膜组织中热休克蛋白(HSP27、HSP70、HSP90)的表达变化。方法将40只SD大鼠随机均分为空白对照组及造模2周组、造模4周组、造模6周组。模型组每日给予4.25%葡萄糖持续非卧床性腹膜透析液(100 m L/kg)腹腔注射,建立腹膜纤维化大鼠模型,空白对照组每日给予等量的生理盐水腹腔注射。各组于末次腹膜透析后48 h行腹膜平衡试验评估腹膜转运功能(超滤量、小分子溶质转运率);采集大鼠腹膜组织,用HE染色法和Masson染色法观察其组织病理学变化;采用免疫组化法检测大鼠腹膜组织TGF-β1、Smad2/3、HSP27、HSP70、HSP90蛋白表达。结果随腹膜透析时间延长,模型组大鼠腹膜超滤量逐渐降低,小分子溶质转运率逐渐升高(P均0.05);腹膜逐渐增厚,间质有炎症细胞浸润伴水肿及毛细血管增生;腹膜基质层增厚,胶原纤维增多,各组大鼠肠系膜组织厚度、腹膜组织厚度比较差异有统计学意义(P均0.05)。各组大鼠腹膜组织TGF-β1、Smad2/3、HSP27、HSP70、HSP90蛋白表达比较差异有统计学意义(P均0.05)。结论成功构建腹膜纤维化大鼠模型;其腹膜组织中HSP27、HSP70、HSP90蛋白呈高表达,且透析时间越长,以上指标表达变化越明显。
[Abstract]:Objective to establish a rat model of peritoneal fibrosis and to investigate the expression of heat shock protein (HSP27) HSP70 (HSP90) in peritoneal tissues of rats. Methods Forty Sprague-Dawley rats were randomly divided into blank control group, 2 week model making group, 4 week model making group and 6 week model making group. The model group was given 4.25% glucose continuously intraperitoneal injection of 100 mL / kg peritoneal dialysate (100 mL / kg) daily to establish the model of peritoneal fibrosis, and the blank control group was given the same amount of normal saline daily intraperitoneal injection. Peritoneal balance test was performed 48 hours after the last peritoneal dialysis to evaluate peritoneal transport function (ultrafiltration, small molecule solute transport rate), peritoneal tissues were collected and histopathological changes were observed by HE staining and Masson staining. The expression of TGF- 尾 1 Smad2 / 3 HSP27HSP70 HSP90 protein in rat peritoneal tissues was detected by immunohistochemical method. Results with the prolongation of peritoneal dialysis time, the peritoneal ultrafiltration decreased gradually, the transport rate of small molecule solute increased gradually, the peritoneal membrane became thicker, the interstitial inflammatory cells infiltrated with edema and capillary hyperplasia, and the peritoneal matrix layer thickened. There were significant differences in the thickness of mesenteric tissue and peritoneal tissue between each group (P < 0.05). There were significant differences in the expression of TGF- 尾 1 Smad2 / 3 HSP27, HSP70, HSP90 and HSP90 in the peritoneal tissues of rats in each group (P < 0.05). Conclusion the rat model of peritoneal fibrosis was successfully constructed, and the expression of HSP27, HSP70, HSP90 was higher in peritoneal tissue, and the longer the dialysis time was, the more obvious the expression of HSP27, HSP70 and HSP90 was.
