解郁安神方对慢性温和应激抑郁动物模型学习记忆功能的影响

【摘要】:目的研究解郁安神方对慢性温和应激(CMS)抑郁动物模型学习记忆功能的影响。方法按照体重将成年雄性昆明小鼠随机分为5组(每组6只):空白组,模型组,大中小3个剂量实验组(解郁安神方,50,100,200 mg·kg~(-1))。制备CMS抑郁大鼠模型。用Morris水迷宫(MWM)评价实验动物空间学习记忆功能。结果给药后第4天,模型组与小中大3个剂量实验组平台潜伏期分别是(42.33±7.76)s与(27.83±6.34),(17.00±6.42),(12.67±5.28)s,与模型组比较,3个剂量实验组差异均有统计学意义(均P0.05),提示解郁安神方醇提物可改善CMS抑郁动物模型学习功能。给药后第5天,模型组与大剂量实验组平台象限潜伏期分别是(48.67±13.37),(24.33±15.33)s,与模型组比较,大剂量实验组差异有统计学意义(P0.05)。模型组与中大2个剂量实验组平台象限时间分别是(38.17±11.36)s与(64.17±8.84),(65.00±9.75)s,与模型组比较,2个剂量实验组差异均有统计学意义(均P0.05),提示解郁安神方醇提物可改善CMS抑郁动物模型记忆功能。结论解郁安神方可以影响CMS抑郁动物模型MWM定位航行训练与空间探索测试不同阶段行为学变化,提示其可改善CMS抑郁动物模型学习记忆功能。
[Abstract]:Objective to study the effect of Jieyu Anshen recipe on learning and memory function of chronic mild stress depression animal model. Methods the adult male Kunming mice were randomly divided into 5 groups according to their body weight (6 mice in each group: blank group, model group, 3 experimental groups with large, medium and small doses) (Jieyu Anshen decoction 50100200 mg / kg). CMS depression rat model was established. The spatial learning and memory function of experimental animals was evaluated by Morris water maze. Results on the 4th day after administration, The plateau latencies of the model group and the small, medium and large dose groups were 42.33 卤7.76 and 27.83 卤6.34, respectively. Compared with the model group, the differences among the three dose groups were statistically significant (P 0.05), suggesting that the alcohol extract of Jieyu Anshen prescription could improve the learning function of CMS depression animal model. On the 5th day after administration, the latencies of platform quadrant in the model group and the experimental group were 48.67 卤13.37 and 24.33 卤15.33 seconds, respectively. Compared with the model group, there was significant difference between the high dose group and the experimental group (P 0.05). The time of platform quadrant in the model group and the middle and large dose groups were 38.17 卤11.36s and 64.17 卤8.84 respectively. Compared with the model group, the differences between the two groups were statistically significant (P 0.05), suggesting that the alcohol extract of Jieyu Anshen prescription could improve the memory function of CMS depression animal model. Conclusion Jieyu Anshen prescription can affect the behavioral changes of MWM positioning navigation training and space exploration test in CMS depression animal model, suggesting that it can improve the learning and memory function of CMS depression animal model.
