大鼠丘脑背外侧核5-羟色胺和5-羟色胺2A受体的表达

【摘要】:大鼠丘脑背外侧核(LD)是丘脑外侧核群的组成部分,与空间学习和记忆有关。其又分为背内侧部(LDDM)和腹外侧部(LDVL)。5-羟色胺(5-HT)作为重要的神经递质与其2A型受体(5-HTR_(2A))也被认为在学习和记忆过程中发挥着非常重要的作用。 目的:本实验应用免疫组化技术,通过观察5-羟色胺及其2A受体在大鼠丘脑背外侧核不同部位的表达情况,为进一步研究丘脑背外侧核、5-羟色胺及其2A受体与学习记忆的关系提供形态学资料。 方法:实验动物采用SD成年大鼠8只,雌雄各半,体重200~250g。由大连医科大学实验动物中心提供。4%水合氯醛腹腔麻醉(1ml/100g体重),仰卧固定,用剪刀剪开胸壁暴露心脏,将灌注针经左心室插至升主动脉。剪开右心耳,快速灌注生理盐水200ml直至右心耳流出液变得无色透明,再灌注4%多聚甲醛固定液(pH7.4)300ml,前100ml快速灌注,后200ml缓慢灌注,持续灌注约2h。灌毕立即取脑,剔净脑表面的脑膜,置于4%多聚甲醛固定液中4℃过夜,然后用振动切片机连续冠状切片,片厚30μm。切取3套,一套用于检测5-HT,一套用于检测5-HTR_(2A),一套做对照实验。采用免疫组织化学染色方法,光镜下观察5-HT及5-HTR_(2A)在丘脑背外侧核不同部位的表达情况,所有标本均使用同一套图像分析系统进行拍照并应用图像分析系统进行分析,所得数据应用SPSS11.5软件进行配对资料t检验。 结果:1. 5-HT免疫组化结果:光镜下,大鼠丘脑背外侧核LDDM和LDVL两部位均可见棕黄色免疫反应阳性的5-HT能神经纤维终末,高倍镜下可见纤维终末走行距离较长,呈串珠状。LDDM和LDVL两部位平均光密度值比较,数值无统计学差异(P0.05)。阴性对照未见阳性表达。2. 5-HTR_(2A)免疫组化结果:在丘脑背外侧核LDDM和LDVL两部位均可见棕黄色的免疫反应阳性神经元,分布均匀,其胞体呈中等大小,免疫反应阳性产物主要定位于神经元胞膜,胞质和胞核不着色。LDDM和LDVL两部位平均灰度值比较,数值无统计学差异(P0.05)。阴性对照未见阳性表达。 结论:在大鼠丘脑背外侧核内存在5-HT能神经纤维终末。5-HTR_(2A)在大鼠丘脑背外侧核内有表达。
[Abstract]:LDD is an integral part of the lateral thalamic nucleus group, which is related to spatial learning and memory. As an important neurotransmitter and its type 2A receptor 5-HTR, LDDM (LDDM) and LDVLA (5-HT) are also considered to play an important role in the process of learning and memory. Objective: to observe the expression of 5-hydroxytryptamine and its 2A receptor in different parts of the dorsal lateral nucleus of the thalamus by immunohistochemical technique. To provide morphological data for further study of the relationship between 5-hydroxytryptamine and its 2A receptor and learning and memory in dorsal lateral thalamic nucleus. Methods: eight adult SD rats, half male and female, were used. An intraperitoneal anaesthesia of 1 ml / 100 g of chloral hydrate was provided by the Experimental Animal Center of Dalian Medical University. After supine fixation, the chest wall was cut open with scissors to expose the heart, and the perfusion needle was inserted into the ascending aorta through the left ventricle. The right atrial appendage was cut open, the normal saline 200ml was infused quickly until the effluent of the right atrial appendage became colorless and transparent, and the pH value of 4% paraformaldehyde fixed solution was 7.4 ~ 300ml after reperfusion. The 100ml was infused rapidly before and after 200ml was infused slowly for about 2 hours.
