兔不同部位脂肪间充质干细胞成骨能力的差异

【摘要】:目的:观察兔不同部位来源的脂肪间充质干细胞(ADSCs)分化成骨的能力。 方法:分别从一只新西兰兔的腹股沟、腹膜后、颈背部取出脂肪组织并提取脂肪间充质干细胞,体外培养传至第5代,加入诱导剂分别进行成骨诱导,不加入诱导剂的为对照组。 结果:经钙钴染色及茜素红染色后,诱导组大部分细胞呈阳性反应,以腹股沟部脂肪间充质干细胞的结节体积最大,对照组未见明显阳性细胞。细胞内碱性磷酸酶(ALP)活性检测表明诱导组比对照组ALP活性明显升高(P<0.05);腹股沟部脂肪间充质干细胞ALP活性较颈背部及腹膜后高,差异有统计学意义(P<0.05);颈背部与腹膜后相比较无明显差异(P>0.05)。 结论:1、兔ADSCs经过体外诱导后,,具有成骨能力。2、腹股沟部脂肪可作为骨组织工程种子细胞来源的较佳取材部位。
[Abstract]:Aim: to observe the ability of adipose mesenchymal stem cells (ADSCs) derived from different parts of rabbit to differentiate into bone. Methods: adipose tissue and adipose mesenchymal stem cells were extracted from the groin and peritoneum of a New Zealand rabbit respectively. The adipose mesenchymal stem cells were cultured to the fifth passage in vitro. The control group did not add the inducer. Results: after calcium cobalt staining and alizarin red staining, most of the cells in the induction group showed positive reaction, and the tubule size of the inguinal adipose mesenchymal stem cells was the largest, but no obvious positive cells were found in the control group. The activity of alkaline phosphatase (ALP) in the induction group was significantly higher than that in the control group (P < 0.05), and the ALP activity in the inguinal adipose mesenchymal stem cells was higher than that in the back of neck and retroperitoneum. The difference was statistically significant (P < 0.05), but there was no significant difference between the back of neck and retroperitoneum (P > 0.05). Conclusion ADSCs has the ability of osteogenesis in vitro. The inguinal fat can be used as the best source of seed cells for bone tissue engineering.
