阪崎肠杆菌新疆分离株的生物学特性及乳鼠器官侵袭特征研究

【摘要】:目的:对阪崎肠杆菌新疆分离株的生物学特性进行分析,通过乳鼠实验分析其器官侵袭特征,探讨可能的毒力因素。方法:用结晶紫中性红胆盐葡萄糖琼脂(VRBGA)酪胨大豆胨琼脂(TSA)、脑心浸液培养液(BHI)、阪崎肠杆菌显色培养基(DFI),酪蛋白氨基酸酵母浸膏培养液(CAYE)培养各菌株,观察不同菌株的生长特征;采用纸片扩散法进行抗生素的药敏试验;阪崎肠杆菌新疆分离株经产毒培养后对3~4日龄ICR乳鼠连续灌饲4日,并于第7日后无菌解剖取肠、肝、脾、肾等组织,石蜡切片,HE染色,光镜观察。结果:在不同培养基中不同菌株的生长特征不完全相同,经CAYE产毒培养液后,菌株表现出两种不同的培养特征,有的产生絮状沉淀伴有贴壁现象,有的则无絮状沉淀且无贴壁现象,在其他培养基中的培养特征无差别;药敏试验表明所有阪崎肠杆菌新疆分离株都对碳青霉烯类抗生素亚胺培南和美洛培南100%耐药,对至少20种抗生素敏感;阪崎肠杆菌新疆分离株经乳鼠实验测得肠尸比均小于0.083,显示肠毒素阴性,但经乳鼠灌饲实验观察及病理切片分析,发现阪崎肠杆菌新疆分离株仍能造成乳鼠生长发育迟缓和器官的损伤,并且这种损伤是全身性的,包括肠、肝、脾、肾等重要器官。结论:阪崎肠杆菌是肠杆菌科耐药性不显著的细菌,阪崎肠杆菌新疆分离株对机体具有潜在危害。
[Abstract]:Objective: to analyze the biological characteristics of Enterobacter sakazakii isolated from Xinjiang, and to explore the possible virulence factors by analyzing the organ invasion characteristics of the isolated strains of Enterobacter sakazakii. Methods: the strains were cultured with crystal violet neutral red bile salt glucose Agar (VRBGA) peptone soybean peptone Agar, brain heart extract (BHI), Enterobacter sakazakii color medium (DFI), casein amino acid yeast extract (CAYE), and the growth characteristics of different strains were observed. Enterobacter sakazakii strains isolated from Xinjiang were inoculated with enterobacter sakazakii for 4 days after inoculation for 4 days, and the tissues of intestine, liver, spleen and kidney were dissected on the 7th day. Paraffin sections were stained with HE and observed under light microscope. Results: the growth characteristics of different strains were not identical in different culture medium. After using CAYE medium, the strains showed two different culture characteristics, some of which produced flocculent precipitates and adhered to the wall. Some of them had no flocculent sedimentation and no adherence to the wall, and there was no difference in the culture characteristics in other media, and the drug sensitivity test showed that all isolates of Enterobacter sakazakii in Xinjiang were resistant to imipenem and melopenem antibiotics, which were 100% resistant to carbapenem antibiotics. Enterobacter sakazakii isolates from Xinjiang were found to be less than 0.083, which were negative for enterotoxin, but were observed by feeding experiment and analyzed by pathological section. It was found that Enterobacter sakazakii isolates from Xinjiang can still cause growth retardation and organ damage in young mice, and the damage is systemic, including important organs such as intestine, liver, spleen, kidney and so on. Conclusion: Enterobacter sakazakii is a bacterium with no significant drug resistance in Enterobacteriaceae, and Enterobacter sakazakii isolates from Xinjiang have potential harm to organism.
