超高精度小鼠彩色断层数据集的建立

【摘要】:背景:小动物成像及影像研究的广泛应用要求实验者掌握小鼠的断面解剖,小鼠鼻腔鼻窦等研究不能通过通常的活体解剖手段来获得其解剖知识。目的:将现有的数字虚拟人技术应用于小鼠,建立一个超高精度的小鼠连续断层彩色数据集。方法:将成年雄性Balb/c小鼠处死后褪毛,改良Kawamoto’s方法进行冰冻包埋,用Leica CM3600XP冰冻切片机以5μm的层厚进行无间隔连续切片,采用2 400万像素Nikon D750单反相机连续拍摄断面图像,以JPG格式储存。参照规则断面法,使用photoshop7软件裁剪包埋块的边缘,并进一步校正图像大小、调整亮度和配准,以TIFF格式保存。将数据集导入Amira 6.0,用最小二乘法进一步配准,直接对小鼠进行面重建及体重建以判断数据集的质量。结果与结论:(1)获取小鼠无间隔连续断面原始图像6 034张,数据量87 GB,经过图像处理和配准,获得184 G小鼠断层数据集,重建了满意的小鼠三维模型;(2)基于研究采用的图像采集系统及处理流程,建立的虚拟小鼠数据集具有分辨率高、还原真实性强的特点,将方便实验小鼠精细解剖的研究,并为相关教学及实验研究提供了重要的工具。
[Abstract]:Background: the extensive application of imaging and imaging of small animals requires the experimenters to master the sectional anatomy of mice, and the study of mouse nasal cavity and nasal sinuses cannot obtain its anatomical knowledge by the usual living anatomical methods. Aim: to establish an ultra-high precision continuous tomographic color data set of mice by applying the existing digital virtual human technology to mice. Methods: after the adult male Balb/c mice were killed, the hair of the adult male Balb/c mice was killed, and the modified Kawamoto's method was used to embed the hair. The Leica CM3600XP frozen slicer was used to slice the sections without interval with a thickness of 5 渭 m. The cross-section images were continuously taken with a 24 million pixel Nikon D750 SLR camera. Store in JPG format. Referring to the regular section method, photoshop7 software is used to cut the edge of the embedded block, and further calibrate the image size, adjust the brightness and registration, and save it in TIFF format. The data set was imported into Amira 6.0 and further registered with the least square method. The face reconstruction and body weight of the mice were carried out directly to judge the quality of the data set. Results and conclusion (1) 6 034 unspaced continuous cross-sectional images were obtained, and 87 GB of data were obtained. After image processing and registration, 184G mouse fault data sets were obtained. Based on the image acquisition system and processing flow, the virtual mouse data set has the characteristics of high resolution and strong reduction authenticity, which will facilitate the study of fine anatomy of experimental mice. It also provides important tools for related teaching and experimental research.
