IL-2或IL-15协同结核分枝杆菌抗原诱导人外周血单个核细胞体外扩增细胞的特性比较

【摘要】:目的分析白细胞介素2(IL-2)或IL-15协同结核分枝杆菌抗原(MTB-Ag)诱导人外周血单个核细胞(PBMC)扩增细胞的生物学特性。方法采用MTB-Ag联合IL-2或MTB-Ag联合IL-15体外分别刺激正常人PBMC,培养至第12天时,经流式细胞术检测扩增细胞中γδT细胞、自然杀伤(NK)细胞和CD4+T细胞所占的比例,以及分析3种淋巴细胞扩增的绝对数量。同时用MTT法检测两组扩增细胞对肿瘤细胞的杀伤活性。结果与MTB-Ag联合IL-2刺激组相比,MTB-Ag联合IL-15刺激组扩增细胞中γδT细胞所占比例明显高于前者,NK细胞所占比例明显低于前者;而两组扩增细胞中CD4~+T细胞所占比例并无显著性差异。与2个刺激组中3种淋巴细胞数量的分析结果一致。并且MTB-Ag联合IL-15刺激组扩增细胞对不同肿瘤细胞均具有显著的杀伤活性,但与MTB-Ag联合IL-2刺激组相比,并无显著性差异。结论 IL-15协同MTB-Ag可高效诱导人PBMC产生以γδT细胞增殖为主的效应细胞,该扩增细胞在体外对肿瘤细胞具有显著的杀伤活性。
[Abstract]:Objective to investigate the biological characteristics of human peripheral blood mononuclear cells (PBMCs) induced by interleukin-2 (IL-2) or IL-15 in combination with Mycobacterium tuberculosis antigen (MTB-Ag). Methods normal PBMCs were stimulated by MTB-Ag combined with IL-2 or MTB-Ag combined with IL-15 in vitro. The percentage of 纬 未 T cells, natural killer NK cells and CD4 T cells were detected by flow cytometry at the 12th day of culture. The absolute number of lymphocyte amplification was analyzed. At the same time, MTT assay was used to detect the cytotoxicity of expanded cells to tumor cells. Results the percentage of 纬 未 T cells in the expanded cells of MTB-Ag + IL-15 group was significantly higher than that of the former group, but there was no significant difference between the two groups in the percentage of CD4T cells. The results were consistent with the results of the analysis of three kinds of lymphocytes in the two stimulation groups. The expanded cells of MTB-Ag combined with IL-15 had significant cytotoxicity to different tumor cells, but there was no significant difference compared with MTB-Ag combined with IL-2. Conclusion IL-15 combined with MTB-Ag can effectively induce the proliferation of 纬 未 T cells in human PBMC, and the expanded cells have significant cytotoxicity to tumor cells in vitro.
