清肺化浊行血法防治PM2.5对慢性阻塞性肺疾病大鼠模型的影响

【摘要】:目的:对雾霾PM2.5建立的慢性阻塞性肺疾病(COPD)大鼠模型可靠性及清肺化浊行血法治疗雾霾形成的COPD疗效作出评价。方法:90只雄性Wistar大鼠随机分为两组:正常对照组(30只)、COPD模型组(60只),采用暗火造成雾霾环境烟熏加气管滴注脂多糖的造模方法建立COPD模型,并将其分为B1、B2、B3、B4组。B1组留取标本,饲养30d后处死;B2、B3、B4组大鼠撤去一切造模因素后正常饲养。B3、B4组分别给予相应药物干预治疗,连续4周,后观察大鼠一般情况、肺功能及血清中白介素-8(IL-8)和白介素-10(IL-10)含量情况,并对其病理标本进行检测。结果:COPD大鼠模型的一般情况与其临床表现相符,大鼠体质量减轻,吸气阻力和呼气阻力升高、肺顺应性下降,但血清IL-8、IL-10浓度与正常组相比,差异显著(P0.05),病理切片显示肺组织出现炎性损伤。用康肺散结化浊行血汤和布地奈德干预的模型大鼠症状好转,肺功能改善,血清IL-10浓度与模型组大鼠相比,显著降低(P0.05),病理切片示炎性损伤明显减轻。结论:用本实验方法建立COPD大鼠模型是可行的,康肺散结化浊行血汤在一定程度上能够改善COPD大鼠模型的症状。
[Abstract]:Objective: to evaluate the reliability of rat model of chronic obstructive pulmonary disease (COPDD) induced by PM2.5 haze and the efficacy of clearing lung and removing turbid blood in treating COPD caused by smog. Methods 90 male Wistar rats were randomly divided into two groups: normal control group (n = 30) and COPD model group (n = 60). The COPD model was established by smoke fumigation and endotracheal instillation of lipopolysaccharide (LPS) in the smog environment caused by dark fire, and the model was divided into two groups: group B _ (1) B _ (2) B _ (2) B _ (3) B _ (4) and group B _ (1). After 30 days of feeding, the rats in group B _ 2 B _ 3 B _ 3H _ 4 were killed and all the model factors were removed. The rats in group B _ (3) B _ (4) were given corresponding drug intervention for 4 weeks respectively. The general condition, lung function and the contents of interleukin-8IL-8 (IL-8) and interleukin-10 (IL-10) in serum of rats were observed. The pathological specimens were examined. Results the general situation of the model was consistent with its clinical manifestation. The weight of rats was reduced, inspiratory resistance and expiratory resistance were increased, and lung compliance was decreased, but the concentration of IL-8 IL-10 in serum was higher than that in normal group. The difference was significant (P 0.05). Pathological sections showed inflammatory injury in lung tissue. The model rats treated with Kangfei Sanjie Huazhuo decoction and budesonide improved their symptoms and pulmonary function. Compared with the model group, the concentration of IL-10 in serum decreased significantly (P 0.05), and the pathological section showed that the inflammatory injury was alleviated obviously. Conclusion: it is feasible to establish COPD rat model by this method. Kangfei Sanjie Huazhuazhuoxue decoction can improve the symptoms of COPD rat model to some extent.
