2014—2015年江苏省肠道病毒71型VP1基因特征分析

【摘要】：目的:探讨2014—2015年江苏省肠道病毒71型(enterovirus 71,EV71)分离株的病毒基因特征。方法:用RD细胞对RT-PCR初筛EV71阳性的标本进行病毒分离,成功分离到的EV71分离株进行VP1区的序列测定和分析。序列分析结果用DNA Star和MEGA 6.0软件进行分析,并与各型参考株进行比较。结果:共分离到35株EV71型毒株,分离阳性率为29.17%。35株EV71分离株VP1区的核苷酸和氨基酸序列同源性分别为93.3%~100.0%和96.6%~100.0%,与EV71的C4a亚型代表株安徽阜阳株的核苷酸和氨基酸序列同源性分别为95.6%~97.9%和97.9%~100.0%;在系统进化树上与C4a亚型代表株相聚在一起,并且存在2个进化分支。结论:2014—2015年江苏省分离的35株EV71毒株均属于C4a亚型;江苏省EV71流行株可能与国内其他省市流行的病毒有着共同的祖先,再各自进化。
[Abstract]:Objective: to study the gene characteristics of enterovirus 71 (EV71) isolated from Jiangsu Province in 2014-2015. Methods: the EV71 positive specimens screened by RT-PCR were isolated by Rd cells, and the EV71 isolates were sequenced and analyzed. The sequence analysis results were analyzed by DNA Star and Mega 6.0 software, and compared with the reference strains. Results: 35 strains of EV71 were isolated. The nucleotide and amino acid sequence homology of the VP1 region of the isolated strain 29.17.35 was 93.3% and 96.6100.0%, respectively. The nucleotide and amino acid sequence homology with the C4a subtype of EV71 in Fuyang strain of Anhui Province was 95.697. 9% and 97. 997. 0% respectively, and the nucleotide and amino acid sequence homology was 97. 990. 0. 00.The nucleotide and amino acid sequence homology of EV71 C4a subtype strain was 95. 6% and 97. 9% respectively. The chemotypic tree was joined with the representative strain of C4a subtype. And there are two evolutionary branches. Conclusion 35 EV71 strains isolated from Jiangsu Province in 2014-2015 belong to C4a subtype, and the EV71 epidemic strains in Jiangsu Province may share a common ancestor with other provinces and cities in China, and then evolve separately.
