高血压脑动脉硬化家兔模型制作

【摘要】:目的(1)寻找一种存活率高且能快速制作出高血压脑动脉硬化模型的方法。(2)探寻家兔脑动脉硬化的早期观察指标。 方法42只新西兰大白兔随机分为7组:A组:对照组,B组:双肾动脉0.70mm狭窄组,C组:右肾切除左肾动脉0.70mm狭窄组,D组:双肾动脉0.55mm狭窄组,E组:右肾切除左肾动脉0.55mm狭窄组,F组:双肾动脉0.45mm狭窄组,G组:右肾切除左肾动脉0.45mm狭窄组。分别于术前(0周)、术后1、2、4、8、12周测血压,术后开始高脂喂养,高脂喂养前(0周)、高脂饲料饲喂4、8、12周测血脂,12周后等压灌注固定,取颈总动脉、胸主动脉、主动脉弓、脑组织做病理检测,并测量脑动脉血管外径(D)、内径(d),计算D/d-1的值。 结果C组、D组各有2只家兔死亡,E组3只家兔死亡,F组、G组全部死亡,其余家兔均完成实验。D组、E组在第1周血压显著升高(P0.01),并持续至实验结束。0周时各组间血脂水平无显著差别(P0.05),4周时至12周时实验组TC、LDL较对照组显著升高(P0.05)。实验组颈总动脉、胸主动脉及主动脉弓均可见颈动脉粥样硬化斑块。脑动脉D/d-1的值:VG染色及actin染色A组与B组无显著差异(P0.05),其余各组间比较P=0.000。 结论1.以高血压并高脂血症制作的家兔模型有颅内动脉硬化的表现。2.D/d-1的值可作为家兔脑动脉硬化的早期观察指标。3.高血压脑动脉硬化模型以双侧肾动脉0.55mm狭窄联合高脂喂养法制作存活率高、血压升高时间早、操作简便。
[Abstract]:Objective 1) to find a method of high survival rate and rapid establishment of hypertensive cerebral arteriosclerosis model. Methods 42 New Zealand white rabbits were randomly divided into 7 groups: control group B: bilateral renal artery 0.70mm stenosis group C group: right nephrectomy left renal artery 0.70mm stenosis group D group: double renal artery 0.55mm stenosis group E group: right nephrectomy left renal artery 0.55mm stenosis group F group: double renal artery 0.45mm stenosis group G group: right nephrectomy left renal artery 0.45mm stenosis group. The blood pressure was measured at 0 weeks before operation, 1 day and 2 minutes after operation. Blood pressure was measured at 12 weeks after operation. After operation, high fat feeding was started, and high fat diet was fed at 0 weeks before feeding. Blood lipids were measured and fixed with isobaric perfusion and fixation after 12 weeks. Common carotid artery, thoracic aorta and aortic arch were taken from the common carotid artery, thoracic aorta and aortic arch. The pathological examination of brain tissue and the measurement of the external diameter of cerebral artery and the internal diameter of the cerebral artery were performed to calculate the value of D / d-1. Results there were 2 rabbits in group C and 2 rabbits in group D, 3 rabbits in group E and 3 rabbits in group F, and all of them died in group G. All the other rabbits completed the experiment. The blood pressure in group E increased significantly at the first week, and there was no significant difference in blood lipid levels between the groups until the end of the experiment. The level of serum lipid in the experimental group was significantly higher than that in the control group from the 4th week to the 12th week after the end of the experiment, and the level of TCfU LDL in the experimental group was significantly higher than that in the control group. Carotid atherosclerotic plaques were found in the common carotid artery, thoracic aorta and aortic arch in the experimental group. There was no significant difference between group A and group B (P 0.05), and there was no significant difference between group A and group B (P 0.05), and there was no significant difference between group A and group B (P 0.05), and the other groups were compared with each other (P = 0.000). Conclusion 1. The model made with hypertension and hyperlipidemia has the manifestation of intracranial arteriosclerosis. 2. The value of D / d-1 can be used as an early observation index of cerebral arteriosclerosis in rabbits. Hypertensive cerebral arteriosclerosis model was made with bilateral renal artery 0.55mm stenosis combined with high fat feeding method with high survival rate, early blood pressure rise time and simple operation.
