静磁场作用下金属(钴、铬)离子对成骨细胞生物学影响的实验研究

【摘要】:目的:探讨静磁场(SMF)作用下金属离子对成骨细胞生物活性的影响,为防治假体周围无菌性松动提供一定的理论依据。 方法:将CoCl2粉末与CrCl3粉末溶于无菌双蒸水配制成CoCl2和CrCl3金属离子混合溶液,将金属离子Co2+、Cr3+混合溶液与小鼠颅骨成骨细胞(MC3T3-E1)共培养,根据实验分组加载不同磁场强度(1、10、100mT)。实验分为Co2++Cr3+组(对照组);Co2+、Cr3++1mT; Co2+、Cr3++10mT; Co2+、Cr3++100mT。扫描电镜检测成骨细胞表面超微结构。MTT法检测成骨细胞活力。流式细胞术检测细胞周期分布。酶联免疫分析(ELISA)法对培养上清液ALP(碱性磷酸酶)含量进行检测。 结果:静磁场作用下的成骨细胞呈现更加成熟的形态学特征,Co2+、Cr3+金属离子对成骨细胞MC3T3的毒性明显降低;同时G2M(分裂期)分布比例明显增加,细胞停滞在GoG1(休眠期)的比率明显减少;与对照组相比,不同场强磁场加载后,成骨细胞碱性磷酸酶分泌明显增强(P0.05)。 结论:静磁场作用下Co2+、Cr3+金属离子对成骨细胞的毒性作用会明显减低,为磁场疗法防治假体周围骨溶解提供了实验依据。
[Abstract]:Aim: to investigate the effect of metal ions on the biological activity of osteoblasts under the action of SMF in order to provide a theoretical basis for the prevention and treatment of aseptic loosening around prosthesis. Methods: CoCl2 powder and CrCl3 powder were dissolved in aseptic double distilled water to form a mixed solution of CoCl2 and CrCl3 metal ions. The mixed solution of CoCl2 and CrCl3 was co-cultured with MC3T3-E1 of mouse skull osteoblasts. The experiment was divided into two groups: Co2Cr3 group (control group), Co2Cr310mTand Co2Cr3100mT. The surface ultrastructure of osteoblasts was examined by scanning electron microscope. MTT assay was used to detect the viability of osteoblasts. Cell cycle distribution was detected by flow cytometry. Enzyme linked immunosorbent assay (Elisa) was used to detect ALP (alkaline phosphatase) in culture supernatant. Results: under the action of static magnetic field, osteoblasts showed more mature morphological characteristics. The toxicity of Co _ 2 / Cr _ 3 to MC3T3 was significantly decreased, and the distribution of G _ 2M (mitotic phase) was significantly increased. Compared with the control group, the alkaline phosphatase secretion of osteoblasts increased significantly after different magnetic fields. Conclusion: the toxicity of Co _ 2 / Cr _ (3) metal ions to osteoblasts can be significantly reduced under the action of static magnetic field, which provides experimental basis for magnetic field therapy to prevent and treat osteolysis around prosthesis.
