裕固族男性饮酒行为与乙醇代谢酶基因ADH3和ALDH2多态型的关系

【摘要】:目的研究裕固族男性饮酒行为及其与相关遗传学特征的关系,为饮酒相关疾病的防治提供科学依据。 方法(1)采用自评式问卷对肃南县505名裕固族男性饮酒状况进行调查,通过单因素和多因素回归分析探讨饮酒行为的影响因素;(2)采用酒精依赖性诊断量表(ADS)筛选出酒精依赖者,比较洒精依赖人群和非酒精依赖人群血脂、肝功和肾功能以及乙醇脱氢酶3(ADH3)和乙醛脱氢酶2(ALDH2)基因多态型的分布。基因多态型的检测采用聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)方法。 结果(1)裕固族男性饮酒率为56.83%,酒精依赖患病率为9.90%;Logistic回归结果提示与饮酒行为有独立相关关系的因素包括家庭收入、婚姻状况、日吸烟量(正相关P0.05)、年龄、文化程度、职业、健康状况(负相关P0.05);与酒精依赖有独立相关关系的因素包括年龄、家庭收入、婚姻状况、日吸烟量、年饮酒量(正相关P0.05)、文化程度、职业、健康状况(负相关P0.05);(2)饮酒量与血脂、肝肾功能指标存在相关关系;(3)ALDH2*2等位基因的分布与饮酒量负相关,裕固族男性ALDH2*1和ALDH2*2等位基因的分布频率分别为:76.82%和33.18%:ADH3*1和ADH3*2等位基因的分布频率分别为:87.87%和16.13%。 结论裕固族男性的饮酒率和酒精依赖的患病率均较高,长期大量饮酒者出现血脂代谢异常、肝功能异常等症状。醇脱氢酶3(ADH3)和醛脱氢酶2(ALDH2)基因多态性在嗜酒的形成中起一定作用,ADH3和ALDH2基因多态性可能对嗜酒的形成具有联合作用。
[Abstract]:Objective to study the relationship between drinking behavior and genetic characteristics of Yugur men, and to provide scientific basis for the prevention and treatment of alcohol-related diseases. Methods 1) A self-assessment questionnaire was used to investigate the drinking status of 505 Yugur men in Sunan County. The influencing factors of drinking behavior were investigated by univariate and multivariate regression analysis. The alcohol-dependent patients were screened by using the Alcohol dependence Diagnostic scale (ADS). The distribution of blood lipids, liver function and renal function, and the polymorphism of alcohol dehydrogenase (ADH3) and acetaldehyde dehydrogenase (ALDH2) gene in spermatozoa dependent population and non-alcohol dependent population were compared. Polymerase chain reaction (PCR)-restriction fragment length polymorphism (RFLP) was used to detect gene polymorphism. Results 1) the alcohol consumption rate of Yugur men was 56.83, and the prevalence of alcohol dependence was 9.90%. Logistic regression analysis showed that the independent correlation factors with drinking behavior included family income, marital status, daily smoking amount (positive correlation P 0.05, age, education, occupation). Health status (negative P0.05); factors with independent correlation with alcohol dependence included age, family income, marital status, daily smoking, annual alcohol consumption (positively correlated P0.05, educational level, occupation), There was a negative correlation between alcohol consumption and blood lipids, liver and kidney function, and the distribution of ALDH2O2 allele in health status (P 0.05), and the distribution of ALDH _ 2O _ 2 allele was negatively correlated with alcohol consumption. The allelic frequencies of ALDH2O1 and ALDH2O2 alleles in Yugur ethnic group were respectively: 76.82% and 33.18%, respectively. The frequencies of alleles of ALDH2O1 and ADH3O2 were: 87.87% and 16.13%, respectively. Conclusion the prevalence of alcohol consumption and alcohol dependence were higher in Yugur nationality men. The abnormal metabolism of blood lipids and liver function were found in a large number of alcohol drinkers for a long time. Alcohol dehydrogenase (ADH3) and aldehyde dehydrogenase (ALDH2) gene polymorphisms may play a role in the formation of alcoholism. ADH3 and ALDH2 gene polymorphisms may play a combined role in the formation of alcoholism.
