乌头和“半蒌贝及”配伍对甲醛致痛模型小鼠镇痛作用的影响

【摘要】:目的:考察“半蒌贝及攻乌”反药组合对制川乌镇痛效果的影响并探索相关机制。方法:基于甲醛足底皮下注射致痛模型,检测致痛小鼠给药后2个时相的舔足时间,明确制川乌和法半夏、栝楼、川贝母、浙贝母、白及各单药及1∶1、1∶2和1∶3配伍组合的镇痛效果,并通过联用κ阿片受体激动剂U-69593或拮抗剂Nor-BNI以及ELISA检测致炎足NO、PGE2和TNF-α含量方法,进一步分析相关作用机制。结果:制川乌单用能明显缩短致痛小鼠2个时相的舔足时间,配伍法半夏、栝楼、浙贝母和白及后此作用减弱,呈量效关系。制川乌联用Nor-BNI后可使舔足时间延长至不给药的状态;联用U-69593后舔足时间比之前缩短,再配伍法半夏、白及后舔足时间明显延长;制川乌单用能明显降低甲醛致痛小鼠炎症足中NO、PGE2和TNF-α的含量,配伍法半夏、栝楼、浙贝母和白及后相关因子含量升高。结论:降低乌头的镇痛功效可能是“半蒌贝及攻乌”反药组合相反的具体表现之一,其机制与干扰κ阿片受体活化及增高炎症局部促炎介质含量有关。
[Abstract]:Aim: to investigate the analgesic effect of the antidrug combination of Phellodendron chinensis and Gongwu on the analgesic effect of Radix Aconitum mongolicum and to explore the related mechanism. Methods: based on the model of pain induced by subcutaneous injection of formaldehyde into plantar, the licking time of two phases after administration of the drug was detected in mice, and the time of licking foot was determined to determine the preparation of Radix Polygoni, Trichosanthes kirilowii, Fritillaria thunbergii and Thunb. The analgesic effects of each single drug and 1: 1: 1: 1: 2 and 1:3 combination were studied, and the related mechanisms were further analyzed by the combination of U-69593 or Nor-BNI, as well as Elisa to detect the levels of NOPGE2 and TNF- 伪 in inflamed feet. Results: the time of lapping feet in two phases of pain induced mice could be shortened obviously by single use of Radix Polygoni. The effect of compatibility of Pinellia ternata, Trichosanthes kirilowii, Fritillaria thunbergii and White was weakened, showing a dose-effect relationship. After using Nor-BNI, the time of licking foot was prolonged to the state of no administration, the time of licking foot was shorter than that before, and the time of licking foot was obviously prolonged after the combination of U-69593 and the method of compatibility with Pinellia ternata, white and after licking. The contents of NONP-GE2 and TNF- 伪 in the inflammatory feet of mice with formaldehyde-induced pain were significantly decreased, and the contents of related factors such as Pinellia ternata, Trichosanthes kirilowii, Fritillaria thunbergii and white were increased. Conclusion: reducing the analgesic effect of aconitum may be one of the opposite manifestations of anti-drug combination of "half Trichosanthes" and "Gongwu", and its mechanism is related to interfering with the activation of 魏 opioid receptor and increasing the content of inflammatory mediators.
