外侧隔核ghrelin对大鼠胃运动的调控及机制研究

【摘要】:目的:探讨外侧隔核ghrelin对大鼠胃运动的调控及机制。 方法:采用核团微量注射、传感器胃内置入及免疫组化研究方法,记录清醒自由活动大鼠胃运动幅度和频率的变化,探讨外侧隔核(lateral septal nucleus, LS)微量注射ghrelin及其受体阻断剂(D-Lys3-GHRP-6)对大鼠胃运动的影响,并观察迷走复合体(DVC)内c-Fos的表达,及双侧膈下迷走神经切断后,LS注射ghrelin对大鼠胃运动的影响。 结果:①外侧隔核分别注射20 nmol,200 nmol或2000 nmol ghrelin 6min后,大鼠胃收缩幅度显著增高(P0.05-0.01),胃收缩频率显著加快(P0.05-0.01),且呈显著剂量依赖关系。外侧隔核注入ghrelin+D-Lys3-GHRP-6混合液6 min后,与ghrelin+NS组比较,ghrelin促进胃收缩幅度(P0.05-0.01)和频率(P0.05)的作用消失,但单用ghrelin受体拮抗剂D-Lys3-GHRP-6对胃收缩幅度(P0.05)和收缩频率(P0.05)均无显著影响;②外侧隔核注射200nmol ghrelin 90 min后,与NS组比较,大鼠迷走复合体内c-Fos表达明显增加(P0.01);③膈下迷走神经切断后,大鼠胃收缩幅度(P0.05)和收缩频率(P0.05)显著降低;在膈下迷走神经切断大鼠,与假手术对照组相比,外侧隔核注射200nmol ghrelin,胃收缩幅度(P0.05-0.01)和收缩频率(P0.05)显著降低,但与迷走神经切断+NS组相比,胃收缩幅度(P0.05)和频率(P0.05)无显著改变。 结论:Ghrelin在外侧隔核通过作用其受体显著促进大鼠胃运动。Ghrelin在外侧隔核促进胃运动的作用可能下行通过迷走复合体-迷走神经通路实现。
[Abstract]:Objective: to investigate the regulation and mechanism of ghrelin in lateral septal nucleus on gastric motility in rats. Methods: the changes of the amplitude and frequency of gastric motility were recorded by microinjection of nucleus, intragastric implantation of sensors and immunohistochemical study in rats with conscious and free movement. To investigate the effect of microinjection of ghrelin and its receptor blocker (D-Lys3-GHRP-6) into the lateral septal nucleus (lateral septal nucleus, LS) on gastric motility in rats, and to observe the expression of c-Fos in vagus complex and the effect of ghrelin injection on gastric motility after bilateral subphrenic vagotomy. Results the gastric contraction amplitude and gastric contraction frequency were significantly increased (P0.05-0.01) and gastric contraction frequency (P0.05-0.01) were increased in a dose-dependent manner after injection of 20 nmol or 2000 nmol ghrelin 6min into the lateral septal nucleus. After injection of ghrelin D-Lys3-GHRP-6 for 6 min, the effects of ghrelin on gastric contraction amplitude (P0.05-0.01) and frequency (P0.05) were disappeared compared with those in ghrelin NS group, but D-Lys3-GHRP-6 alone had no significant effect on gastric contractile amplitude (P0.05) and contractile frequency (P0.05). 2After the injection of 200nmol ghrelin 90 min into the lateral septal nucleus, compared with the NS group, the expression of c-Fos increased significantly (P0.01) after subphrenic vagotomy, the gastric contraction amplitude (P0.05) and the contractile frequency (P0.05) were significantly decreased. After subphrenic vagotomy, the amplitude of gastric contraction (P0.05-0.01) and the frequency of gastric contraction (P0.05) were significantly decreased after injection of 200nmol ghrelin in the lateral septal nucleus compared with the sham operated control group, but the amplitude of gastric contraction (P0.05) and the frequency of gastric contraction (P0.05) were not significantly changed compared with the NS group. Conclusion the effects of the receptor in the lateral septal nucleus on the gastric motility of rat. Ghrelin in the lateral septal nucleus may be mediated by vagus pathway via vagus pathway.
