【摘要】:目的:本研究旨在探讨IFN-β和angiostatin分别在离体和在体条件下对淋巴管内皮细胞生成的影响。 方法:1、淋巴管在体抑制试验:观察活体兔被切断的淋巴管应用抑制因子后的再生情况。2、猪胸导管内皮细胞的培养及形态学观察:应用光镜、同焦显微镜、透射电镜观察离体胸导管内皮细胞的形态结构。3、淋巴管内皮细胞离体抑制试验:应用划线法和MTT法来确定IFN-β对淋巴管内皮细胞生成的抑制作用。4、细胞调亡检测:采用Hoechst法检测细胞凋亡。 结果:1、手术切断兔前或后肢淋巴管5天后,生理盐水侧(对照组)的淋巴管已愈合;angiostatin侧(试验组)仍有大量染料渗漏。2、对培养的淋巴管内皮细胞进行了形态学观察。3、划线法和MTT法均显示IFN-β对离体淋巴管内皮细胞的增殖和游走有明显的抑制作用。4、IFN-β可引起细胞凋亡 结论:1、angiostatin对在体淋巴管的愈合有抑制作用。2、IFN-β对离体淋巴管内皮细胞的增殖和游走有明显的抑制作用,且有剂量依赖性。3、IFN-β具有促进细胞凋亡的作用。
[Abstract]:Aim: to investigate the effects of IFN- 尾 and angiostatin on lymphatic endothelial cell formation in vitro and in vivo, respectively. Methods: in vivo inhibition test of lymphatic vessels in vivo: to observe the regeneration of lymphatic vessels in vivo after the application of inhibitory factor in rabbits, and to observe the culture and morphology of endothelial cells in porcine thoracic ducts: using light microscope, isokinetic microscope, Transmission electron microscopy (TEM) was used to observe the morphological structure of thoracic ductal endothelial cells in vitro. The inhibitory effect of IFN- 尾 on lymphatic endothelial cells was determined by the method of underline and MTT, and the apoptosis of lymphatic endothelial cells was detected. Apoptosis was detected by Hoechst method. Results the lymphatic vessels in the saline side (the control group) were healed 5 days after the surgical transection of the lymphatic vessels in the anterior or hind limbs of the rabbits. There was still a large amount of dye leakage in the angiostatin side (experimental group). Morphological observation of cultured lymphatic endothelial cells (LECs) was carried out. The results showed that IFN- 尾 could inhibit the proliferation and migration of lymphatic endothelial cells in vitro. Conclusion: (1) angiostatin can inhibit the healing of lymphatic vessels in vivo. [WT5HZ] IFN- 尾 can inhibit the proliferation and migration of lymphatic endothelial cells in vitro. Moreover, IFN- 尾 can promote apoptosis in a dose-dependent manner.
