HIF-1α、TGF-β1在肺低氧代谢中的表达及作用机制研究

【摘要】:第一章HIF-1α、IL-8在肺低氧代谢中的表达 目的研究肺低氧代谢中血清中缺氧诱导因子1α(HIF-1α),促炎细胞因子白介素-8(IL-8)表达,探讨低氧代谢与其相关性。 方法40只SD大鼠随机分为对照组(A组),实验组:30min单肺低氧组(B组),60min单肺低氧组(C组),120min单肺低氧组(D组);A组采用气管切开插管行双肺通气,实验组行气管切开插管行单肺通气,建立AHPI模型,分别于实验30min、60min、120min取大鼠血液及非通气侧肺组织,通过透射电镜检查非通气侧肺组织细胞结构变化,采用ELISA法,Western blot法测定HIF-1α,IL-8含量。 结果A组镜下可见AEC-Ⅱ的核质均匀,可见特有的L(blamellar body板层小体)。B组AEC-Ⅱ微绒毛(Microvilli Mv)排列混乱,Lb排空略微增强;C组Lb数目增加,排空增强;D组核固缩,出现巨大的Lb,伴空泡样变。实验组肺组织及血清中HIF-1α的蛋白含量明显高于对照组且随低氧代谢时间延长呈上升趋势:ABCD (P0.01),而IL-8的含量与对照组相比却无明显差异(P0.05)。 结论低氧代谢致大鼠肺组织细胞结构发生改变,HIF-1α的表达上调,且与低氧代谢时间呈正相关性。 第二章TGF-β1、AIF在肺低氧代谢中的表达 目的研究肺低氧代谢中肺泡上皮Ⅱ型细胞(alveolar epithelial cells type II,AEC-Ⅱ)的超微结构变化与肺组织及血清中转化生长因子β1(Transforming growth factor-β1,TGF-β1)和凋亡诱导因子(apoptosis inducing factor ,AIF)表达,探讨其之间关系。 方法40只SD大鼠随机均分为A,B,C,D组(n=10),A、B、C组采用气管切开插管行单肺通气,建立AHPI模型,D组采用气管切开插管行双肺通气。分别于实验30min(A)、60min(B)、120min(C,D)取大鼠血液,离心取血清冻存,取左下肺组织待透射电镜检查和蛋白含量测定。用ELISA法,Western blot法分别测定在血清和肺组织的TGF-β1、AIF表达。 结果A组AEC-Ⅱ微绒毛(Microvilli Mv)排列混乱,Lb数目增多,排空略微增强。B组Lb数目增加,密度明显降低,排空增强,部分呈空泡样变。C组核固缩,未见核仁,出现巨大的Lb,伴空泡样变。D组镜下可见AEC-Ⅱ的细胞核,核质均匀,胞质内可见AEC-Ⅱ特有的Lb(lamellar body板层小体),无空泡样变及排空现象。A、B、C组肺组织与血清中TGF-β1、AIF的相对含量明显高于D组且随缺氧时间延长呈上升趋势:DABC (P0.01)。 结论低氧代谢致大鼠肺组织细胞结构发生改变,TGF-β1和AIF的含量在试验时间内表达呈上调趋势。
[Abstract]:Chapter 1 expression of HIF-1 伪 and IL-8 in hypoxic Metabolism objective to study the expression of hypoxia inducible factor-1 伪 (HIF-1 伪) and pro-inflammatory cytokine interleukin-8 (IL-8) in serum of pulmonary hypoxia metabolism, and to explore the correlation between hypoxia metabolism and hypoxia metabolism. Methods Forty Sprague-Dawley rats were randomly divided into two groups: control group (group A), control group (group A, n = 30 min), group B (group B, n = 60), group A (n = 120 min), group A (n = 120), group A were treated with tracheotomy and intubation, and group A, were treated by tracheotomy and intubation, respectively. The AHPI model was established. The blood and non-ventilated lung tissues of the rats were collected at 30min or 60min for 120min, respectively. The changes of cellular structure in the non-ventilated lung tissues were examined by transmission electron microscopy. The content of HIF-1 伪 -IL-8 was measured by ELISA method.
