应激大鼠肠道树突细胞的免疫功能

【摘要】:目的研究应激大鼠肠道树突细胞(DC)表型及功能状态的改变。方法选用20只雄性SD大鼠随机分为对照组、实验组,各10只。给予实验组大鼠结直肠扩张联合束缚刺激,对照组大鼠不处理,采用大鼠腹部收缩反射(AWR)对两组大鼠进行直肠气囊扩张,测量肠道容量感觉阈值。成功建立模型后,磁珠分选技术分离肠系膜淋巴结DC,检测其表面CD80、CD86、MHCⅠ、MHCⅡ、Toll样受体(TLR)4的表达,将MLNDC与脾脏单个核细胞体外混合培养,流式标记CD4+/CD8+T淋巴细胞,检测MLNDC促CD4+/CD8+T淋巴细胞增殖的能力。结果实验组大鼠的疼痛阈值明显低于对照组(P0.05);实验组大鼠MLNDC表达MHCⅡ水平高于对照组(P0.05),而表达CD80、CD86、MHCI与对照组比较差异无统计学意义(P0.05);实验组大鼠MLNDC表达TLR4高于对照组(P0.05);与对照组相比,实验组大鼠MLNDC促CD4+/CD8+T淋巴细胞增殖能力增强(P0.05)。结论大鼠应激刺激后肠道DC上调表达TLR4,刺激其本身分化成熟,进一步表达MHCⅡ,后者通过介导T淋巴细胞活化,引起肠道低度炎症,从而引起肠道内脏高敏感的产生。
[Abstract]:Objective to study the changes of (DC) phenotype and function of intestinal dendritic cells in stressed rats. Methods Twenty male SD rats were randomly divided into control group (n = 10) and experimental group (n = 10). Rats in the experimental group were given colorectal dilatation combined with bound stimulation, while the control group rats were not treated. The rats in the two groups were dilated by abdominal contractile reflex (AWR), and the sensory threshold of intestinal volume was measured.
