洋苷菊正丁醇提取物对哮喘模型小鼠的作用机理

【摘要】:目的研究维药洋苷菊提取物对支气管哮喘小鼠模型的治疗作用,探讨其抗哮喘的可能机制。方法 KM种小鼠60只随机分为6组:正常对照组,模型组,阳性对照组,洋苷菊正丁醇提取物低、中、高剂量组。采用卵蛋白(OVA)腹腔注射致敏与雾化吸入激发制作哮喘模型;致敏第15天开始每次雾化激发前1 h给予洋苷菊治疗,连续4周。末次给药后1 h,检测各组小鼠的肺功能及增强的呼吸间歇;取肺泡灌洗液(BALF)测定细胞计数,颈动脉取血,离心取血清测定白介素IL-2、IL-10及IL-12含有量;肺组织匀浆离心取上清液检测超氧化物歧化酶(SOD)和丙二醛(MDA)水平;HE染色观察肺组织病理改变。结果洋甘菊正丁醇提取物可改善哮喘小鼠肺功能;与正常对照组相比,模型组增强的呼吸间歇(Penh)明显升高(P0.01);与模型组相比,各给药组Penh显著降低小鼠(P0.05或P0.01),BALF嗜酸性粒细胞计数(EOS)及肺组织MDA各组水平均有一定程度的降低(P0.05或P0.01);各治疗组肺组织SOD及血清中IL-2、IL-10、IL-12含有量明显升高(P0.05,P0.01)。结论洋甘菊正丁醇提取物具有明显的平喘作用;洋甘菊正丁醇提取物对肺泡灌洗液中EOS、肺组织MDA水平有一定降低作用;对血清IL-2、IL-10、IL-12及肺组织SOD水平有一定升高作用;洋甘菊正丁醇提取物通过改善哮喘小鼠肺功能,降低Penh,改善哮喘小鼠气道高反应性。
[Abstract]:Objective to study the therapeutic effect of the extract of guerarin on asthma model in mice, and to explore the possible mechanism of its anti-asthma. Methods 60 km mice were randomly divided into 6 groups: normal control group, model group, positive control group, low, middle and high dose group. The asthmatic model was induced by intraperitoneal injection of ovalbumin (OVA) and inhalation of atomization, and the rats were treated with guanosin 1 hour before each atomization on the 15th day of sensitization for 4 weeks. At 1 hour after the last administration, lung function and enhanced respiratory interval were detected in each group, cell count was determined by (BALF) in alveolar lavage fluid, blood was taken from carotid artery, IL-10 and IL-12 content in serum were measured by centrifugation. Superoxide dismutase (SOD) and malondialdehyde (MDA) (MDA) were detected by HE staining in supernatant of lung tissue homogenate. Results the pulmonary function of asthmatic mice was improved by n-butanol extract from chamomile, the (Penh) of the model group was significantly higher than that of the normal control group (P0.01), and that of the model group was higher than that of the model group. The eosinophilic granulocyte count (EOS) and the level of MDA in lung tissue were significantly decreased in each treatment group (P0.05 or P0.01), and the contents of IL-2IL-10 IL-12 in lung tissue and serum of each treatment group were significantly increased (P0.05, P0.01). Conclusion the n-butanol extract of Chamomile has obvious antiasthmatic effect, the extract of n-butanol can decrease the level of MDA in alveolar lavage fluid, increase the level of IL-10 IL-12 in serum and SOD in lung tissue. Chamomile n-butanol extract improved airway hyperresponsiveness of asthmatic mice by improving lung function, reducing Penhs and improving airway hyperresponsiveness of asthmatic mice.
