磁场对成纤维细胞的影响及量效关系研究

【摘要】:近年来随着非药物疗法与保健的兴起,磁疗因具有操作简单、无创无痛的特点,而越来越受到人们的关注,对磁场生物效应的报道也越来越多。 但是目前磁疗基础研究薄弱,临床上无严格使用规范,磁对生物体作用的量效关系和安全性研究尚不够深入,磁对生物体作用的内在机制不清楚。本研究在对磁疗用永磁磁源空间磁场分析的基础上,对正常培养和缺氧培养条件下的成纤维细胞受磁场影响的结果进行研究,分析了磁场对成纤维细胞作用的量效关系,探讨了磁场对生物体的作用机制。本文的主要研究内容包括: 1.分析磁疗用圆片永磁磁源的空间磁感强度分布,设计体外细胞培养加磁方法,准确掌握磁场作用于细胞的磁感强度。 2.通过MTT法检测细胞的活性,研究磁场对成纤维细胞增殖活性的影响。实验发现,选取的不同磁感强度磁场均不同程度上提高了成纤维细胞的活性。 3.通过测细胞培养液上清中LDH的含量,研究磁场对成纤维细胞膜损伤的影响。结果发现,实验中选取的大部分磁源对成纤维细胞膜是安全的。 4.通过测量细胞培养液上清中SOD与MDA的含量,研究了不同强度永磁磁场对细胞氧化损伤的影响。结果显示为,细胞的抗氧化性经过磁场的作用后被加强了。但氧化损伤的产物有多有少。 5.利用流式细胞仪检测磁场对成纤维细胞凋亡的影响,从实验结果来看,不同强度永磁磁场对成纤维细胞凋亡的影响显现出了磁场对细胞的阈值作用。但大多磁场组对于成纤维细胞凋亡的影响并无规律可循。 本文研究不同磁感强度永磁磁源对成纤维细胞的影响,从细胞水平探讨磁场的定量作用效果和量效关系,为规范永磁磁源的使用和磁作用效果的研究提供基础性实验依据。明确提出磁场的有效作用磁感强度量值或磁感强度量值范围、以及磁场安全使用的磁感强度量值。
[Abstract]:In recent years, with the rise of non-drug therapy and health care, magnetic therapy has been paid more and more attention because of its simple operation, non-invasive and painless characteristics, and there are more and more reports on the biological effects of magnetic field. However, at present, the basic research of magnetic therapy is weak, there is no strict standard in clinical use, the study on the dose-effect relationship and safety of magnetic action on organism is not deep enough, and the internal mechanism of magnetic action on organism is not clear. Based on the spatial magnetic field analysis of permanent magnetic source for magnetic therapy, the effects of magnetic field on fibroblasts in normal and anoxic culture were studied, and the dose-effect relationship of magnetic field on fibroblasts was analyzed. The mechanism of magnetic field acting on organism is discussed. The main contents of this paper are as follows: 1. The spatial magnetic intensity distribution of magnetic disk magnetic source for magnetic therapy was analyzed, and the method of in vitro cell culture and magnetization was designed to accurately grasp the magnetic intensity of magnetic field acting on cells. 2. The effect of magnetic field on the proliferation of fibroblasts was studied by MTT assay. It was found that the activity of fibroblasts was increased by magnetic field with different intensity of magnetic inductance. The effect of magnetic field on the damage of fibroblast cell membrane was studied by measuring the content of LDH in the supernatant of cell culture medium. The results showed that most of the magnetic sources selected in the experiment were safe for fibroblast cell membrane. 4. 4. By measuring the content of SOD and MDA in the supernatant of cell culture medium, the effect of permanent magnetic field on cell oxidative damage was studied. The results showed that the antioxidant activity of cells was enhanced by magnetic field. But the products of oxidative damage are less. 5. The effect of magnetic field on the apoptosis of fibroblasts was detected by flow cytometry. From the experimental results, the effect of magnetic field on the apoptosis of fibroblasts showed the threshold effect of magnetic field. However, the effect of most magnetic field groups on fibroblast apoptosis is not regular. In this paper, the effects of permanent magnetic sources with different magnetic intensity on fibroblasts are studied, and the quantitative effect and dose-effect relationship of magnetic field are discussed from the cell level, which provides a basic experimental basis for standardizing the use of permanent magnetic sources and the study of magnetic effects. It is clear that the effective action of magnetic field or the range of magnetic inductance intensity, as well as the safe use of magnetic field.
