Wistar大鼠腮腺放射损伤模型建立

【摘要】:目的模拟临床分割放疗下的腮腺放射损伤,建立大鼠腮腺放射损伤后的放射性病理模型,为腮腺放射损伤的相关研究奠定基础。方法选取成年Wistar大鼠20只,随即分为两组;实验组15只使用直线加速器照射,每次5Gy,共40Gy。对照组5只不做放射,正常饲养。观察两组实验动物的饮食体重及腮腺区局部变化;照射完成4周后处死动物,并在光镜观察腮腺组织病理改变。结果放射后实验大鼠均未出现可评价的异常神经行为及体征;腺体组织结构疏松,腺小叶间隙扩大;间质纤维组织增多,血管扩张,片状炎细胞浸润;以浆细胞为主,可见淋巴细胞;胞浆浓缩,染色深,核缩小,深染;腮腺内毛细血管内皮增厚,血管肿胀,周围组织有明显纤维化;血管内皮增生,细胞呈梭形,向血管腔内突出。实验组毛细血管密度明显低于对照组。结论本实验条件下大鼠直线加速器分割照射腮腺,共40Gy,造成了较为典型的唾液腺放射损伤表现,成功的模拟头颈部放疗所致的临床及组织病理学变化,可作为唾液腺放疗损伤模型。
[Abstract]:Objective to simulate the radiation injury of parotid gland induced by clinical fractionation radiotherapy and to establish the radiation-pathological model of parotid gland radiation injury in rats, so as to lay a foundation for the study of radiation injury of parotid gland. Methods Twenty adult Wistar rats were randomly divided into two groups: the experimental group (n = 15) was irradiated with a linear accelerator at a dose of 5 Gy for a total of 40 Gy. 5 rats in the control group were fed normally without radiation. The weight of diet and local changes of parotid gland were observed in the two groups, and the pathological changes of parotid tissue were observed under light microscope after 4 weeks of irradiation. Results No evaluable abnormal neurobehavioral and physical signs were found in the experimental rats after radiation. The glandular structure was loose, the glandular lobular space was enlarged, the interstitial fibrous tissue was increased, vasodilation and inflammatory cells were infiltrated, and plasma cells were the main. Visible lymphocytes; cytoplasm concentration, deep staining, nuclear shrinking, deep staining; parotid gland capillary endothelial thickening, vascular swelling, obvious fibrosis in surrounding tissues; vascular endothelial proliferation, cells spindle shape, protruding into the vascular lumen. The capillary density in the experimental group was significantly lower than that in the control group. Conclusion in this experiment, the parotid gland was irradiated by linear accelerator fractionation with 40 Gy, which resulted in the typical radiation injury of salivary gland. The clinical and histopathological changes of the parotid gland induced by simulated head and neck radiotherapy were successful. It can be used as a model of salivary gland radiation injury.
