建立大鼠福氏志贺菌感染模型及双歧杆菌治疗效果与机制的研究

【摘要】:目的: 1.建立福氏志贺菌感染的SD大鼠模型,为进一步研究志贺菌的致病机制和疫苗防治提供基础。 2.探讨两歧双歧杆菌对福氏志贺菌感染大鼠的治疗效果。 方法: 1.先用硫酸链霉素给正常大鼠灌胃,出现菌群失调症状后,再通过福氏志贺菌灌胃感染大鼠,根据临床表现和病理切片确定感染模型的建立。 2.使用两歧双歧杆菌干预治疗福氏志贺菌感染大鼠模型,检测治疗组中治疗期间大鼠粪便的福氏志贺菌含量变化。 3.检测干预治疗后各组大鼠血中IL-2、肠粘液中sIgA的含量。 4.用MTT法检测双歧杆菌干预治疗对大鼠脾细胞增殖的影响,并用脾脏指数及胸腺指数评价双歧杆菌对SD大鼠的免疫增强作用。 结果: 1.感染大鼠表现为粪质较稀,病理切片显示肠粘膜坏死脱落,表明感染模型建立成功。 2.双歧杆菌治疗后,粪便中志贺菌含量下降迅速。 3.与未干预组和生理盐水组相比双歧杆菌治疗组IL-2、肠粘液中sIgA的水平明显升高。 4. MTT法表明双歧杆菌治疗组志贺菌抗原刺激的脾细胞增殖水平显著高于未治疗组。 结论: 1.成功建立了福氏志贺菌感染的大鼠模型。 2.两歧双歧杆菌治疗能使感染模型粪便中志贺菌含量迅速下降。 3.两歧双歧杆菌对福氏志贺菌感染机体的IL-2、sIgA分泌有促进作用。 4.双歧杆菌能促进感染机体的脾细胞增殖。
[Abstract]:Objective: 1. The SD rat model of Shigella flexneri infection was established to provide a basis for further study on the pathogenic mechanism and vaccine control of Shigella flexneri. To investigate the therapeutic effect of bifidobacterium on rats infected with Shigella flexneri. Methods: 1. The normal rats were given streptomycin sulfate first and then infected by Shigella flexneri. The infection model was established according to the clinical manifestations and pathological sections. 2. The rat model of Shigella flexneri infection was treated with bifidobacterium. The content of Shigella flexneri in feces of rats in the treatment group was measured during the treatment period. 3. The content of sIgA in IL-2, intestinal mucus of rats in each group after intervention was detected. 4. 4. The effects of Bifidobacterium on the proliferation of spleen cells in rats were detected by MTT method. The immune enhancement of SD rats was evaluated by spleen index and thymus index. Results: 1. The pathological sections showed intestinal mucosal necrosis and shedding, indicating that the infection model was established successfully. 2. After Bifidobacterium treatment, Shigella content in feces decreased rapidly. The level of sIgA in intestinal mucus of IL-2, in Bifidobacterium treated group was significantly higher than that in non-intervention group and normal saline group. 4. 4. MTT assay showed that the proliferation level of spleen cells stimulated by Shigella antigen in Bifidobacterium treated group was significantly higher than that in untreated group. Conclusion: 1. The rat model of Shigella flexneri infection was established successfully. Bifidobacterium bifidobacterium treatment can rapidly reduce Shigella content in feces of infected model. Bifidobacterium can promote the secretion of IL-2,sIgA in Shigella flexneri infection. 4. Bifidobacterium can promote the proliferation of spleen cells infected with the body.
