鸟苷酸结合蛋白2调控β-葡聚糖诱导的小鼠树突状细胞成熟

【摘要】:目的研究鸟苷酸结合蛋白2(GBP2)对树突状细胞(DC)成熟和功能的影响。方法通过基因芯片和实时定量PCR研究β-葡聚糖诱导的DC基因的变化情况,转染GBP2小干扰RNA(si RNA),敲低GBP2的表达,流式细胞术检测DC表面CD11c、主要组织相容性复合体Ⅱ(MHC-Ⅱ)、CD80水平,ELISA检测细胞上清中白细胞介素6(IL-6)、IL-12p70、TNF-α和IL-10水平。T细胞增殖实验检测全葡聚糖颗粒(WGP)刺激的成熟DC在转染GBP2-si RNA后,对卵清蛋白(OVA)特异性CD4+T细胞的促增殖效应。结果β-葡聚糖诱导后,DC的GBP2 m RNA水平显著降低;敲低DC的GBP2水平后,其表面CD11c、主要组织相容性复合体Ⅱ(MHC-Ⅱ)、CD80水平降低,同时,IL-6、IL-12、TNF-α分泌下降;经WGP刺激的成熟DC在转染GBP2-si RNA后,对OVA特异性CD4+T细胞的促增殖效应降低。结论 GBP2能调控β-葡聚糖诱导的DC成熟,进一步抑制T细胞的增殖。
