脂联素对老化骨髓间充质干细胞自发性骨化的影响

【摘要】:目的探讨老化骨髓间充质干细胞(BMSCs)的自发性骨化过程及脂联素在这一过程中的作用。方法将第2代BMSCs(分离自一小儿畸形断肢的残指)分为空白对照组及脂联素低、中、高剂量组。空白对照组加入DMEM/F12培养基及10%胎牛血清,脂联素低、中、高剂量组分别在此基础上加入终浓度为0.5、1.0、2.0μg/m L的脂联素,各组均传代培养至第10代。收集各组第2代和第10代细胞,茜素红S染色后计算钙化物染色率,q PCR法检测骨钙素(OCN)mRNA相对表达量。结果与第2代细胞比较,各组第10代细胞钙化物染色率、OCN mRNA相对表达量均升高(P均0.05);与空白对照组第10代细胞比较,脂联素低、中、高剂量组钙化物染色率、OCN mRNA相对表达量均升高,但脂联素中、高剂量组升高更明显(P均0.05)。结论 BMSCs在传代过程中可发生自发性骨化,脂联素可促进这一过程,并呈浓度依赖性。
[Abstract]:Objective to investigate the spontaneous ossification of aging bone marrow mesenchymal stem cells (BMSCs) and the role of adiponectin in the process. Methods the second generation of BMSCs was divided into blank control group and adiponectin low, middle and high dose groups. In the blank control group, adiponectin was added to DMEM/F12 medium and 10% fetal bovine serum, and adiponectin was added to 0.51.0 渭 g / mL adiponectin at the final concentration of 0.51.0 渭 g / mL in the middle and high dose groups respectively. The adiponectin was subcultured to the 10th generation in each group. The second and tenth passage cells of each group were collected and the relative expression of osteocalcin (OCN) mRNA was detected by, q PCR method after alizarin red S staining. Results compared with the second passage cells, the relative expression of, OCN mRNA in the 10th passage cells was significantly higher than that in the second passage cells, and the adiponectin level was lower than that in the control group (P < 0.05), and the level of adiponectin was lower than that in the control group (P < 0.05). The relative expression of, OCN mRNA in high dose group was higher than that in high dose group (P 0. 05), but adiponectin in high dose group was higher than that in high dose group (P 0. 05). Conclusion spontaneous ossification may occur in the passage of BMSCs, and adiponectin can promote this process in a concentration dependent manner.
