靶向干扰前列腺癌抗原-1表达对前列腺癌裸鼠移植瘤的影响

【摘要】:目的 观察靶向干扰前列腺癌抗原-1(PCA-1)表达对前列腺癌裸鼠移植瘤的影响。 方法 培养人前列腺癌LNCaP细胞并将其注射到裸鼠皮下建立前列腺癌裸鼠移植瘤模型。将裸鼠随机分为实验组和对照组,实验组:将靶向干扰PCA-1表达的重组质粒PSMAe/p-PCA-1-pA-Sliencer注射到裸鼠移植瘤局部和周围,对照组:将PBS注射到移植瘤局部和周围;观察移植瘤生长情况和实验结束时肿瘤的大小。应用免疫组织化学方法和常规病理技术观察肿瘤组织中PCA-1表达情况以及裸鼠主要器官的病理变化。 结果 实验组裸鼠移植瘤生长速度明显慢于对照组,差异具有统计学意义(P0.05);实验结束时实验组和对照组的移植瘤体积(cm3)分别为1.142±0.157和3.086±0.265,差异有统计学意义(P0.05)。实验组裸鼠的主要器官未出现明显病理变化。实验组裸鼠移植瘤组织PCA-1蛋白染色平均总积分(3.8±1.48)高于对照组(6.2±2.48),差异具有统计学意义(P0.05)。 结论 成功建立前列腺癌裸鼠移植瘤模型,靶向干扰PCA-1表达可以抑制移植瘤的生长,对裸鼠主要器官未产生明显影响。
[Abstract]:Objective to investigate the effect of target interfering prostate cancer antigen 1 (PCA-1) expression on transplanted prostate cancer in nude mice. Methods Human prostate cancer LNCaP cells were cultured and injected subcutaneously into nude mice to establish transplanted tumor model of prostate cancer in nude mice. The nude mice were randomly divided into experimental group and control group. In the experimental group, the recombinant plasmid PSMAe/p-PCA-1-pA-Sliencer, which targeted to interfere with the expression of PCA-1, was injected into and around the transplanted tumor of nude mice. In the control group, PBS was injected into and around the transplanted tumor. The growth of transplanted tumor and the size of tumor at the end of the experiment were observed. The expression of PCA-1 in tumor tissues and the pathological changes of the main organs of nude mice were observed by immunohistochemical method and routine pathological technique. Results the tumor growth rate of nude mice in the experimental group was significantly slower than that in the control group (P0.05), the cm3 of the experimental group and the control group were 1.142 卤0.157 and 3.086 卤0.265 respectively at the end of the experiment (P0.05). There were no obvious pathological changes in the main organs of the nude mice in the experimental group. The mean total score of PCA-1 protein staining in nude mice transplanted in the experimental group (3.8 卤1.48) was significantly higher than that in the control group (6.2 卤2.48) (P0.05). Conclusion the transplanted tumor model of prostate cancer was successfully established in nude mice. The targeted interference of PCA-1 expression could inhibit the growth of transplanted tumor, but had no obvious effect on the main organs of nude mice.
