空肠弯曲菌致急性周围神经病巴马香猪模型初步建立

【摘要】:目的:建立空肠弯曲菌(Campylobacter jejuni,Cj)感染致急性周围神经病巴马香猪模型,初步探讨以空肠弯曲菌为感染源引起继发免疫相关疾病的发病模式,为研究急性运动感觉性轴索性神经病(acute motor sensory axonal neuropathy, AMSAN)的发病机制、其病理生理及空肠弯曲菌的生长特性、感染特性奠定研究模型基础。 方法:以吉兰-巴雷综合征(Guillain-Barre syndrome,GBS)患者分离培养空肠弯曲菌,经中国疾病预防控制中心鉴定为空肠弯曲菌菌株,并经美国疾病预防控制中心鉴定血清型为penner O:19型。用哥伦比亚血培养基于42℃,10%CO2、5%O2、85%N2培养空肠弯曲菌。纯化菌种制备1×106菌悬液灌喂巴马香猪,灌喂前禁食12小时、禁水4小时。分别于灌喂前后分离、鉴定胃肠道菌群。自灌喂当日起每日观察巴马香猪的临床症状:饮食、大便及肢体活动情况。对出现肢体活动障碍巴马香猪自出现症状7天内取材其脊神经根、坐骨神经及臂丛神经,进行饿酸染色、镀银染色、Luxol-fast-blue染色、免疫组织化学、透射电子显微镜进行神经病理观察。 结果:空肠弯曲菌自然感染巴马香猪后,模型动物于1周内出现腹胀、腹泻,2周左右出现肢体活动障碍;神经病理检查可观察到微管、微丝减少,轴索节段性萎缩,髓鞘示节段性回缩,未见脱髓鞘样变性。神经病理学初步诊断为急性运动、感觉轴索性神经病。 结论:空肠弯曲菌感染巴马香猪可致急性周围神经病,巴马香猪可以作为AMSAN研究的模型动物。初步探讨了神经病理检查方法。
[Abstract]:Objective: to establish a Bama pig model of acute peripheral neuropathy caused by campylobacter jejuni (Campylobacter jejuni,Cj) infection and to explore the pathogenesis of secondary immune-related diseases caused by Campylobacter jejuni as the source of infection. In order to study the pathogenesis of acute motor sensory axonal neuropathy (acute motor sensory axonal neuropathy, AMSAN), its pathophysiology, the growth characteristics of Campylobacter jejuni and the infection characteristics, the model was established.
