结核分枝杆菌靶抗原Rv1789刺激结核感染人群IFN-γ水平研究

【摘要】:目的检测并比较Rv1789刺激结核分枝杆菌(Mtb)感染人群和健康对照者外周血IFN-γ水平。方法以原核表达体系构建、表达、纯化rRv1789。从南京市疾病预防控制中心选择39位受试者,参考临床结核病诊断标准和rCM-WBIA诊断标准将其分为TB感染组(包括结核病患者和潜伏感染者)和健康对照组,进行rRv1789-WBIA试验。结果成功构建pET30b-Rv1789质粒,SDS-PAGE鉴定表达产物分子质量单位为44ku,与预期相符;Western blot证实该蛋白能被相应抗体识别。rRv1789刺激Mtb感染组产生的IFN-γ水平显著高于健康对照组(P0.01);rCM和rRv1789刺激受检者全血IFN-γ水平具有显著相关性(r=0.616,P0.01)。结论 PE/PPE家族蛋白Rv1789具有抗原性,可用作Mtb感染快速诊断和结核疫苗候选抗原。
[Abstract]:Objective to detect and compare the levels of IFN- 纬 in peripheral blood of Rv1789 stimulated Mycobacterium tuberculosis (Mtb) infected population and healthy controls. Methods rRv1789. was constructed, expressed and purified by prokaryotic expression system. Thirty nine subjects were selected from Nanjing Centers for Disease Control and Prevention. They were divided into TB infection group (including tuberculosis patients and latent infections) and healthy control group according to the diagnostic criteria of clinical tuberculosis and rCM-WBIA. The rRv1789-WBIA test was carried out. Results the pET30b-Rv1789 plasmid was successfully constructed, and the molecular weight unit of the expressed product was 44ku. which was consistent with the expectation. Western blot confirmed that the protein could be recognized by the corresponding antibody. The level of IFN- 纬 in Mtb infected group stimulated by rRv1789 was significantly higher than that in healthy control group (P0.01). There was a significant correlation between rCM and rRv1789 stimulated whole blood IFN- 纬 levels (r = 0.616, P 0.01). Conclusion PE/PPE family protein Rv1789 has antigenicity and can be used for rapid diagnosis of Mtb infection and candidate antigen for tuberculosis vaccine.
