芋螺毒素GI抗血清制备及中和活性研究

【摘要】:目的测定芋螺毒素GI与牛血清白蛋白(BSA)偶联物免疫小鼠后产生的抗血清是否具有中和GI毒素作用。方法戊二醛法制备GI-BSA偶联物,免疫小鼠,制备GI小鼠抗血清,应用抗血清的抗体中和实验,验证其对GI的解毒作用。结果 SDS-PAGE蛋白电泳结果显示,GI与BSA成功偶联,GI-BSA在标志物相对分子质量(120×10~3)处有两条新蛋白带;GI-BSA(99μg/只)每隔2周免疫1次,共免疫4次,第4次免疫后第10天抗血清中和滴度效价1∶64 000;混合100及200μl小鼠抗血清与≥1 LD_(50)剂量GI,腹腔注射,100μl血清可使1×LD_(50)毒素(16.3μg/kg)组小鼠75%存活,200μl血清可使25.8μg/kg毒素组小鼠75%存活。结论基于GI-BSA半抗原免疫的小鼠抗血清对GI毒素具有明显的解毒作用。
[Abstract]:Objective to determine whether the antiserum produced by the conjugate of conotoxin GI and bovine serum albumin (BSA) (BSA) can neutralize GI toxin in mice. Methods GI-BSA conjugate was prepared by glutaraldehyde method, mice were immunized and anti-serum of GI mice was prepared. The antiserum neutralization test was used to verify the detoxification of GI. Results SDS-PAGE protein electrophoresis showed that GI was successfully coupled with BSA, and GI-BSA had two new protein bands at the relative molecular weight of the marker (120 脳 10 ~ 3). GI-BSA (99 渭 g / mouse) was immunized once every 2 weeks for 4 times, and the neutralizing titer of antiserum was 1:64 000 on the 10th day after the fourth immunization.
