补阳还五汤联合MSCs移植对TIMP-1干预后MCAO模型大鼠凝血Ⅷ因子的影响

【摘要】:目的:揭示补阳还五汤联合MSCs移植对大脑中动脉栓塞再灌注大鼠模型((MCAO))抗凝血、促进血管新生的机理。方法:采用TIMP-1对模型进行干预,侧脑室给予MSCs,联合补阳还五汤灌胃,采用免疫组化法检测凝血因子Ⅷ的表达。结果:联合补阳还五汤联合MSCs移植组能够降低缺血区凝血因子Ⅷ数量,12 h、24 h最为明显,与模型组、抑制剂组、单纯MSCs组相比,有极显著性差异(P0.01);至36 h时有一定升高,与其他组相比,无显著性差异(P0.05),随后表达Ⅷ因子数量又开始下降,至48 h时,与模型组、抑制剂组相比,有显著性差异(P0.05),与单纯MSCs组相比,无显著性差异(P0.05)。结论:补阳还五汤联合MSC移植,能够降低TIMP-1干预后脑缺血模型大鼠缺血脑组织Ⅷ因子数量,起到抗凝血,抑制血栓的形成。
