鼠伤寒沙门氏菌对肠道屏障的破坏作用
[Abstract]:In order to study the effect of Salmonella typhimurium on the intestinal barrier, the colonic pathological section of mice was used to study the effect of Salmonella typhimurium on intestinal barrier. Blue dextran 2000 osmotic assay combined with transwell cell layer model and fluorescence quantitative PCR method was used to explore the possible mechanism of Salmonella typhimurium destroying intestinal barrier. The results of pathological section showed that the intestinal epithelial cells were damaged by Salmonella typhimurium infection, and the osmotic capacity of blue dextran 2000 in the small intestine of Babl/c mice was increased at 2h and 6h (P0.05). The osmotic capacity of Kunming mice increased significantly at 2 and 4 h (P0.005). The transmembrane resistance was decreased (P0.05) and the mRNA transcription level of intercellular tight junction protein (ZO-1,Claudin-1) gene was down-regulated (P0.005). The results showed that Salmonella typhimurium could destroy the tight junctional structure of the cells by down-regulating the expression of tight junction protein in intestinal epithelial cells, thus destroying the intestinal barrier. The effect of Salmonella typhimurium infection on intestinal barrier was clarified, which provided a basis for the study of pathogenic mechanism of Salmonella typhimurium.
