Keap1-Nrf2通路与线粒体关系的研究进展

【摘要】:当机体受到亲电子物质或活性氧刺激时,核转录相关因子2(nuclear factor erythroid-2 related factor 2,Nrf2)能诱导出一系列的保护性蛋白,从而抵抗内源性或外源性氧化应激造成的损害。越来越多的研究发现,某些组织比如心脏受到氧化应激时,Nrf2的活性和线粒体生物合成有关。在Surfeit 1蛋白(Surfeit locus protein 1,Surf1)突变的小鼠心脏中发现,由于细胞色素C氧化酶的缺乏导致细胞呼吸减少从而激活Nrf2。本文从Kelch样环氧氯丙烷相关蛋白1(Kelch-like ECH-associated protein-1,Keap1)-Nrf2通路与线粒体的关系以及氧化应激条件下Nrf2信号通路与线粒体之间的信号交联为重点进行了综述。
[Abstract]:When an organism is stimulated by an electron-friendly substance or reactive oxygen species (Ros), nuclear transcription-associated factor 2 (nuclear factor erythroid-2 related factor-2) can induce a series of protective proteins, thus resisting the damage caused by endogenous or exogenous oxidative stress. More and more studies have found that Nrf2 activity is associated with mitochondrial biosynthesis in certain tissues such as the heart under oxidative stress. In the heart of mice with Surfeit 1 protein (Surfeit locus protein 1, Surf1) mutation, it was found that the lack of cytochrome C oxidase (cytochrome C oxidase) results in the reduction of cellular respiration and activation of Nrf2.. The relationship between Kelch-like epichlorohydrin-associated protein 1 (Kelch-like ECH-associated protein-1,Keap1)-Nrf2 pathway and mitochondria and the signal cross-linking between Nrf2 signaling pathway and mitochondria under oxidative stress were reviewed in this paper.
