番茄红素对神经病理性疼痛模型大鼠脊髓组织中疼痛相关分子表达的影响

【摘要】:目的:分析番茄红素对神经病理性疼痛模型大鼠脊髓组织中疼痛相关分子表达的影响。方法:取健康雌性SD大鼠30只,根据文献方法建立神经病理性疼痛模型,包括假手术组、模型对照组、模型治疗组各10只。将大鼠处死后获取脊髓L2-L6段,测定其中疼痛相关指标的血清水平、基因及蛋白表达情况。结果:假手术组大鼠的血清IL-17、HMGB-1、Aβ、Tau、C3水平低于模型对照组及模型治疗组,CGRP水平高于模型对照组及模型治疗组,模型治疗组大鼠的血清IL-17、HMGB-1、Aβ、Tau、C3水平低于模型对照组,CGRP水平高于模型对照组(P0.05);假手术组大鼠的ERK、CREB、BDNF、NMDA、AMPA、c-fos mRNA表达量低于模型对照组及模型治疗组,模型治疗组大鼠的脊髓ERK、CREB、BDNF、NMDA、AMPA、c-fos mRNA表达量低于模型对照组大鼠(P0.05);假手术组大鼠的TRPV1、NF-κB、NOS、GFAP、ERK、CREB蛋白表达量低于模型对照组及模型治疗组,Reg表达量高于模型对照组及模型治疗组(P0.05),模型治疗组大鼠的TRPV1、NF-κB、NOS、GFAP、ERK、CREB蛋白表达量低于模型对照组,Reg表达量高于模型对照组(P0.05)。结论:番茄红素可以有效减少神经病理性疼痛模型大鼠的促疼痛基因表达,有望成为日后神经病理性疼痛治疗的新手段。
[Abstract]:Aim: to analyze the effect of lycopene on the expression of pain-related molecules in spinal cord of neuropathic pain model rats. Methods: thirty healthy female SD rats were used to establish neuropathic pain models, including sham operation group, model control group and model treatment group (n = 10). After the rats were killed, the L2-L6 segment of spinal cord was obtained and the serum level, gene and protein expression of pain-related indexes were measured. Results: the levels of serum IL-17,HMGB-1,A 尾 and Tau,C3 in sham operation group were lower than those in model control group and model treatment group, and the level of CGRP was higher than that in model control group and model treatment group. The serum IL-17,HMGB-1, level in model treatment group was higher than that in model control group. The levels of A 尾 and Tau,C3 were lower and the level of CGRP was higher than that of the model control group (P0.05). The expression of ERK,CREB,BDNF,NMDA,AMPA,c-fos mRNA in sham operation group was lower than that in model control group and model treatment group, and the spinal cord ERK,CREB,BDNF,NMDA,AMPA, expression in model treatment group was lower than that in model control group. The expression of c-fos mRNA was lower than that of the model control group (P0.05). The expression of TRPV1,NF- 魏 B and NOS,GFAP,ERK,CREB protein in sham operation group was lower than that in model control group and model treatment group, and the expression of Reg was higher than model control group and model treatment group (P0.05). The expression of TRPV1,NF- 魏 B in model treatment group was higher than that in model control group and model treatment group. The expression of NOS,GFAP,ERK,CREB protein was lower than that of model control group, and the expression of Reg was higher than model control group (P0.05). Conclusion: lycopene can effectively reduce the expression of pain-promoting gene in neuropathic pain model rats, and may become a new method for the treatment of neuropathic pain in the future.
