血清和糖皮质激素调节蛋白激酶1在CNS疾病中的作用研究新进展

【摘要】:血清和糖皮质激素调节蛋白激酶1(SGK1)是1993年在受糖皮质激素刺激的大鼠乳腺癌细胞中发现的一个丝氨酸/苏氨酸蛋白激酶。与多数蛋白激酶不同的是,血清和糖皮质激素调节蛋白激酶1不仅受翻译后的磷酸化/去磷酸化调节,而且存在转录水平上的快速调节机制。它具有广泛的生理功能,如调节离子通道,介导细胞增殖、分化、存活及凋亡的信号转导等。它与帕金森病、阿尔茨海默病等多种中枢神经系统疾病密切相关,在神经元变性、凋亡过程中起着重要作用。本文将从血清和糖皮质激素调节蛋白激酶1的结构、生理功能、与中枢神经系统疾病的关系及其抑制剂等方面的新进展作一综述。
[Abstract]:Serum and glucocorticoid regulated protein kinase 1 (SGK1) is a serine / threonine protein kinase found in rat breast cancer cells stimulated by glucocorticoid in 1993. Unlike most protein kinases, serum and glucocorticoid regulated protein kinase 1 is not only regulated by post-translation phosphorylation / dephosphorylation, but also by a rapid regulatory mechanism at the transcriptional level. It has a wide range of physiological functions, such as regulating ion channels, mediating cell proliferation, differentiation, survival and apoptosis signal transduction and so on. It is closely related to Parkinson's disease, Alzheimer's disease and other central nervous system diseases, and plays an important role in neuronal degeneration and apoptosis. In this paper, the structure and physiological function of serum and glucocorticoid regulated protein kinase 1, the relationship between serum and glucocorticoid regulated protein kinase 1 and central nervous system diseases and their inhibitors are reviewed.
