高温致鼠神经管畸形神经上皮中N-cadherin、bFGF、PCNA的表达及其意义研究

【摘要】:目的:研究高温对鼠胚神经管发育时期神经上皮细胞的粘附聚集、增殖凋亡及细胞生长因子的影响,从而探讨高温致神经管畸形的发生机理。方法:将孕鼠随机分成为实验组30只和对照组25只,实验组鼠于孕8d下午15:30进行42℃水浴20min,对照组鼠置于37℃水浴箱20min。实验组和对照组各10只孕鼠于孕15d处死剖腹取胎,辨认和计数活胎、死胎、吸收胎和畸胎。其余孕鼠35只,分别于37℃和42°C水浴后2h、8h、16h、24h、48h处死,剖腹取胎,置于10%的中性福尔马林液中固定,常规石蜡包埋、连续切片。用免疫组织化学法和图像分析仪检测N-cadherin、bFGF、PCNA在各期实验组和对照组鼠胚神经管上皮细胞的表达变化。结果:①实验组鼠胚神经管畸形率达70.91%,对照组致畸率为0。②N-cadherin的免疫组织化学结果:水浴后2-8h N-cadherin在对照组及实验组神经管上皮细胞表达呈阳性,二者之间的OD值差异无统计学意义,内外界膜、脊索及神经管周围间充质细胞也可见阳性反应。水浴后16-24h, N-cadherin在对照组神经管上皮细胞表达呈强阳性,而在实验组表达呈弱阳性,OD值差异有统计学意义(p0.01),周围组织阳性变化不明显。至水浴后48h实验组与对照组相比,神经上皮N-cadherin免疫阳性反应差异不明显,N-cadherin蛋白在两组神经管组织表达均减弱。③)bFGF的免疫组织化学结果:水浴后2h、8h、16h bFGF蛋白在实验组和对照组神经上皮中表达,两组OD值差异无统计学意义,水浴后24h-48h实验组神经上皮的表达弱于对照组,且OD值差异有统计学意义(p0.05), bFGF在两组神经管上皮的表达随胚龄增加有增强的趋势,内外界膜、脊索和周围间充质也呈现阳性反应。④孕8d, PCNA蛋白广泛表达在神经管组织,神经管上皮细胞、神经嵴细胞、周围间充质细胞均呈现高表达状态。水浴后2h和8h, PCNA在实验组神经组织的阳性表达数低于对照组,但差异无统计学意义,高温处理后16h、24h和48h PCNA在实验组神经管上皮组织的阳性细胞数低于对照组,且差异有统计学意义(p0.05)。水浴处理24h之后,PCNA在实验组和对照组神经组织中的阳性细胞数均有明显下降的趋势。结论:高温可使N-cadherin、bFGF、PCNA在神经管上皮细胞的表达降低,影响细胞间的识别、粘附,干扰细胞增殖与凋亡的动态平衡,诱导NTDs发生,是高温导致神经管缺陷的重要途径之一。
