新疆乌恰县人体泡球蚴线粒体DNA分析

【摘要】：目的通过线粒体DNA测序初步确定新疆乌恰县多房棘球蚴的来源。方法新疆维吾尔自治区南疆乌恰县泡球蚴病患者5例,采集其病灶组织,提取DNA,PCR扩增多房棘球绦虫线粒体DNA Nad2序列,并与已发表的泡球蚴线粒体DNA Nad2序列进行序列比对和系统进化分析。结果新疆南疆乌恰县是一个新的泡型包虫病流行区。Nad2基因序列比对分析及遗传距离矩阵显示,5例AE患者病灶扩增的Em Nad2基因存在差异,人体感染的虫株之间存在低的遗传变异。Nad2基因系统进化树分析表明,这些虫株是亚洲多房棘球绦虫亚型,包含3个单倍体型。结论新疆乌恰县多房棘球绦虫流行株属于亚洲基因型,至少有3种单倍体型。关于乌恰县多房棘球绦虫终末宿主和中间宿主的种群以及其在疾病的传播中的作用需要进一步证实。
[Abstract]:Objective to determine the source of Echinococcus multilocularis in Wuqia County, Xinjiang by mtDNA sequencing. Methods five patients with alveolar echinococcosis in Wuqia County, South Xinjiang Uygur Autonomous region, were enrolled in this study. The lesion tissues were collected and the mtDNA sequences of Echinococcus aeruginosa were extracted by DNA,PCR, and the sequence alignment and systematic evolution analysis were carried out with the published mtDNA sequences of Echinococcus aeruginosa. Results Wuqia County, southern Xinjiang, was a new epidemic area of vesicular echinococcosis. Nad2 gene sequence alignment analysis and genetic distance matrix showed that there were differences in Em Nad2 gene amplified in 5 patients with AE, and there was low genetic variation among human infected strains. Nad2 gene phylogenetic tree analysis showed that these strains were Echinococcus multilocularis subtypes in Asia, including three haplotypes. Conclusion the epidemic strain of Echinococcus multilocularis in Wuqia County of Xinjiang belongs to Asian genotype and has at least three haplotypes. The population of terminal host and intermediate host of Echinococcus multilocularis and its role in the spread of disease need to be further confirmed.
