MRTF-A调控miR-1273g-3p对Aβ 25-35 损伤的SH-SY5Y细胞自噬的影响及机制研究

目的:在Aβ25-35诱导人源性SH-SY5Y损伤细胞上,筛选能差异化表达并与MRTF-A相关的miRNA,探讨MRTF-A介导miRNA对Aβ25-35诱导的SH-SY5Y细胞自噬的影响及机制。方法:1.在Aβ25-35诱导SH-SY5Y损伤细胞模型中,用基因芯片筛选与MRTF-A调控相关的miRNA:建立细胞模型;细胞分组(control,Aβ25-35,MRTF-A,MRTF-A+Aβ25-35)。转染MRTF-A质粒48h后,给予40μmol/L Aβ25-35处理,24h后,用western blot方法检测MRTF-A的蛋白表达水平,并提取RNA。用Agilent RNA 6000 Nano/Pico Assay法检测各组RNA质量合格后,使用miRNA One Array芯片技术分析各组中发生显著差异变化的miRNA。得到各组显著变化的miRNA,再通过各组间相互比较得到MRTF-A调控的miRNA。筛选出差异性表达最为显著的miRNA,并用...
