氧化偶氮甲烷-硫酸葡聚糖硫酸钠诱导小鼠结直肠肿瘤方法的优化(英文)

自氧化偶氮甲烷(AOM)-硫酸葡聚糖硫酸钠(DSS)诱导肿瘤用于研究炎症相关或散发性结直肠癌(CRC)以来,数种AOM、DSS处理方式被报导。本文优化了AOM-DSS诱导C57BL/6小鼠散发性结直肠癌模型的方法。利用100μl消毒注射器按照需要剂量一次性腹腔注射适当浓度AOM,DSS处理7天并每天更换新鲜的DSS液体。我们共诱导超过100只小鼠,AOM处理后8至20周时,发现每只小鼠均被诱导发生结直肠肿瘤。肿瘤分布以结直肠远端居多,与人类结直肠癌相似。采用此方法建模的每只小鼠的成瘤数量较多、鼠间肿瘤数量差异较小。此AOM-DSS模型中的肿瘤病变涵盖了瘤前病变、腺瘤性病变、腺瘤以及腺癌。本改良的AOM-DSS结直肠癌建模方法具有简单、经济、成瘤率高等优点。
