【摘要】:固醇调节元件结合蛋白(Sterol Regulatory Element Binding proteins,SREBPs)是一类固定于内质网和核被膜上的膜连接蛋白,具有“碱性螺旋-环-螺旋-亮氨酸拉链”(basic Helix-Loop-Helix-Leucine Zipper,bHLH-Zip)结构,属于bHLH-Zip超家族的一员。已经确认的SREBPs成员有3种:SREBP-1a,SREBP-1c,SREBP-2。其中SREBP-1a,SREBP-1c主要调节脂肪酸(fatty acid synthase,FAS)、甘油三酯(triglyceride,TG)和葡萄糖代谢相关酶基因的表达,SREBP-2主要调控胆固醇代谢。SREBPs在内质网合成后是无功能的前体,与SREBPs裂解激活蛋白(SREBP Cleavage Activating Protein,SCAP)组成复合物,共同调控胞内胆固醇浓度的动态平衡。SCAP含有一个固醇敏感区,是哺乳动物脂质合成和摄入的中心调节因子。SREBP-1c又称脂肪细胞定向与分化因子1(Adipocyte Determination and Differentiation factor-1,ADD1),在脂肪细胞的分化中发挥重要作用。通过过氧化物酶体增殖蛋白激活受体(Peroxisome Proliferator-Activated Receptor,PPARs)和CCAAT元件增强结合蛋白(CCAAT/enhancer binding protein,C/EBPs)共同调控脂肪细胞的分化。SREBPs还参与脂肪合成基因的营养调控,并受胰岛素/葡萄糖和瘦素等多种激素的调控,而且介导胰岛素对多种基因表达的调控。最近研究发现,SREBP-1c的过度表达与肝脏和胰岛等非脂肪组织的脂质积聚相关,是代谢综合征中重要的基因调控连接点。对其调控作用进行全面深入的研究,将对代谢综合征的发病机理和指导临床治疗有更新更全面的认识。
[Abstract]:Sterol regulatory element binding protein (Sterol Regulatory Element Binding proteins, SREBPs) is a kind of fixed on the endoplasmic reticulum and nuclear envelope membrane protein on the connection, with the basic helix loop helix leucine zipper "(basic Helix-Loop-Helix-Leucine Zipper, bHLH-Zip), a member of bHLH-Zip superfamily have been identified. SREBPs there are 3 members: SREBP-1a, SREBP-1c, SREBP-2. and SREBP-1a, SREBP-1c mainly regulates fatty acid (fatty acid synthase, FAS), triglycerides (triglyceride, TG) expression and glucose metabolism related enzyme gene, SREBP-2 mainly regulates the cholesterol metabolism in.SREBPs is a precursor to non functional synthesized in endoplasmic reticulum, and cleavage of SREBPs protein (SREBP Cleavage Activating Protein, SCAP) composed of complex, dynamic balance of.SCAP cholesterol concentration of intracellular co regulation contains a sterol sensitive area, is a mammal Lipid synthesis and intake center regulator.SREBP-1c also known as adipocyte determination and differentiation factor 1 (Adipocyte Determination and Differentiation factor-1, ADD1), play an important role in adipocyte differentiation. Through peroxisome proliferator activated receptor protein (Peroxisome Proliferator-Activated, Receptor, PPARs) and CCAAT enhancer binding protein (CCAAT/enhancer binding element protein, C/EBPs) nutritional regulation of common regulation of adipocyte differentiation of.SREBPs is also involved in lipid synthesis genes, and regulated by insulin / glucose and leptin and other hormones, but also mediated the effect of insulin on the expression of various genes. Recent studies suggest that the overexpression of SREBP-1c and lipid accumulation in liver and pancreatic islet and non fat tissue is related to metabolic syndrome gene regulation important syndrome connection point.
