【摘要】:自从1992年Agre Peter教授第一次从人红细胞膜中分离和鉴定出水通道蛋白1(aquaporin 1,AQP1)以来,后续研究证明水通道蛋白是一个大家族,其成员广泛分布于动物、植物和微生物体内,介导水分子的快速跨膜转运,促进细胞膜水渗透性增加;同时发挥调节细胞容积和细胞内渗透压以及内环境稳态的重要生理作用,这一发现被誉为生命科学研究的一个里程碑,Agre Peter教授因此荣获2003年诺贝尔化学奖;生殖生理和生殖病理的研究一直是人们关注的生殖领域热点问题;生殖过程的许多方面涉及到液体的改变,如卵泡的生长发育、胚泡的发育、精子的发生成熟、精液的形成以及子宫内膜和子宫腔液的周期性变化,均与液体的分泌和吸收有着密切的关系;研究证实有多种水通道蛋白丰富地表达于睾丸、精子、卵泡、子宫、胎盘、胎膜等组织,因此,研究水通道蛋白在生殖系统的表达、作用及其调节机制具有重要的生理学意义;然而,水通道蛋白是否存在于人的子宫内膜中?若存在,其表达特性如何?是否与月经周期存在一定的关系?是否受到卵巢甾体激素的调节?这一系列问题至今未见报道。本研究旨在探讨水通道蛋白在人月经周期子宫内膜中的分布特征和表达规律,并通过体外刺激实验研究雌孕激素对人子宫内膜上皮细胞水通道蛋白2(aquaporin 2,AQP2)表达的调节,以探讨AQP2在人不同月经周期子宫内膜上表达的变化规律及其调节机制。
[Abstract]:Since 1992, Agre professor Peter first isolated from human erythrocyte membrane and identification of water channel protein 1 (aquaporin 1, AQP1), subsequent studies show that water channel protein is a big family, whose members are widely distributed in animal, plants and microorganisms, rapid transmembrane transport mediated by water molecules, promote cell membrane water permeability increased; at the same time play an important physiological role of osmotic pressure and homeostasis regulation of cell volume and intracellular, this discovery is a milepost known as life science research, Professor Peter Agre won the 2003 Nobel prize in chemistry; research on reproductive physiology and reproductive pathology has been the focus that people pay close attention to reproductive field many aspects of the reproductive process; related to fluid change, such as follicular development, blastocyst development, spermatogenesis, sperm formation and endometrial and uterine cavity The periodic change of the liquid, and the liquid absorption and secretion are closely related; research shows that many water channel protein abundantly expressed in testis, sperm, follicle, uterus, placenta, fetal membranes and other organizations, therefore, the research of water channel protein expression in the reproductive system, as well as adjustment mechanism has important physiological significance; however, the water channel protein exists in human endometrium? If there is, its expression characteristics? Whether there is a certain relationship with the menstrual cycle? Is regulated by ovarian steroid hormone? This series of questions has not been reported. This study aimed to investigate the distribution of water channel protein in human endometrium during menstrual cycle and the expression of the law, and through the experimental study of in vitro stimulation of estrogen and progesterone on endometrial epithelial cells of aquaporin 2 (aquaporin 2, AQP2) regulating the expression of AQP2 in different people, to explore Changes in the expression of endometrium on the menstrual cycle and its regulatory mechanism.
