含多表位艾滋病病毒基因的重组脊灰病毒的构建及其免疫研究

【摘要】:由人免疫缺陷病毒(Human immunodeficiency virus,HIV)引起的人获得性免疫缺陷综合症(Acquired Immunodeficiency Syndrome,AIDS,简称艾滋病),已经成为危害人类健康最严重的传染性疾病。目前尚无安全有效的药物可以完全治愈此病,这意味着90%以上的确诊病人最终会死于艾滋病及并发症。虽然目前已有一些抗病毒的药物,但耐药株的出现,复杂的服药过程,昂贵的费用,,以及不能彻底清除病毒等原因,因此艾滋病的最终控制仍有赖于安全有效的疫苗。 目前研制的HIV疫苗种类很多,既有传统的病毒灭活疫苗、减毒活疫苗,也有新型的核酸疫苗、合成肽链疫苗、病毒载体疫苗等,但在临床实验方面效果均不理想。原因可能与HIV的高度变异、致病机理尚不清楚、抗体滴度不高多种原因有关。 脊髓灰质炎病毒是一类单股正链RNA病毒,具有自主的复制能力。减毒的Sabin Ⅰ株疫苗在防止脊髓灰质炎多年的实践中正式安全、有效。所以我们在本实验中采用减毒的Sabin Ⅰ为载体,以HIV中国主要流行株的基因为抗原,研制一种有实际推广价值的防治AIDS的疫苗。 本项研究的目的是基于我国HIV-1主要流行毒株之一B’/C重组株CN54,构建表达HIV基因的多价重组脊髓灰质炎病毒,免疫TgPVR21小鼠,对其免疫效果及免疫机制进行研究探讨。以B’/C重组株CN54的全基因组克隆pCRScript-CN54为模板,PCR扩增gagpold,gp120基因。gagpold和gp120基因通过PCR与启动子串连起来,克隆入转移载体pNeoTK75H6,与脊髓灰质炎病毒共转染P815细胞。经Budr加压选择,
[Abstract]:By the human immunodeficiency virus (Human immunodeficiency virus, HIV) caused by human acquired immunodeficiency syndrome (Acquired Immunodeficiency Syndrome AIDS, referred to as AIDS), has become the most serious infectious diseases endangering human health. There is no safe and effective drugs can cure the disease, which means that more than 90% of patients will die of AIDS and complications. Although there have been some antiviral drugs, but the emergence of resistant strains, taking complex, expensive, and can not completely remove the virus. Therefore, the ultimate control of AIDS still depends on the safe and effective vaccine.
Many kinds of HIV vaccines are developed, both traditional virus inactivated vaccine, live attenuated vaccine, also a new nucleic acid vaccine, synthetic peptide vaccine, virus vector vaccines, but the effect in clinical trials are not ideal. The reason may be highly variant with HIV, the pathogenic mechanism is not clear, the relevant antibody the titer is not high for many reasons.
The polio virus is a single strand RNA virus with self replication. Sabin strain vaccine virus is effective in preventing formal security practice polio for many years. So we use attenuated Sabin 1 as the carrier in this experiment, the HIV Chinese main flow line gene as antigen. To develop a practical preventive AIDS vaccine.
The purpose of this study is based on China's HIV-1 is one of the main epidemic strain B / C recombinant CN54 strain, construct a multivalent recombinant poliovirus HIV gene expression, immune TgPVR21 mice to study the immunogenicity and immune mechanism.
