【摘要】:1996年,Sonoda(Cancer,1996)等建立了一种子宫腺癌细胞系SiSo,他们将SiSo免疫小鼠,利用杂交瘤技术制备了一系列单克隆抗体,发现了一种可被22-1-1单抗识别的新蛋白,分子量为40kD和78kD(认为78KD蛋白为40KD蛋白的二聚体),命名为22—1—1抗原,该抗原不同于以往发现的YH206,GA733,CA125,CEA等肿瘤抗原,认为是一种新的肿瘤相关抗原。Watanabe(Mol Cell Biol,1998)等在1998年从人乳腺癌细胞系MCF-7 cDNA文库中克隆到一个雌激素反应性的新基因,命名为EBAG9(Estrogen receptor-binding fragment-associated gene 9),编码区为639bp,在5'上游有一个雌激素反应元件,该分子mRNA和蛋白的表达随雌激素的诱导而上调。Nakashima(Nat Med,1999)等分离鉴定出编码22—1—1抗原的基因命名为RCAS1(receptor-binding cancer antigen expressed on SiSo cells),基因序列比对后发现RCAS1同EBAG9实际上是同一种蛋白质。 关于RCAS1分子的大多数研究是用22-1-1单抗通过免疫组化实验分析在各种临床肿瘤组织标本中(如宫颈癌、侵袭性宫颈鳞状上皮癌、乳腺癌、T3胃癌等)的表达,认为RCAS1的表达与肿瘤的分化、侵袭性和恶性程度呈正相关,并提示不良预后,RCAS1在正常组织中没有表达。RCAS1可作为肿瘤恶性程度
[Abstract]:In 1996, Sonoda (Cancer, 1996) a uterine adenocarcinoma cell line SiSo, they will SiSo immunized mice were prepared by hybridoma technique for a series of monoclonal antibodies, found a new protein can be identified by mAb 22-1-1, molecular weight of 40kD and 78kD (think 78KD 40KD protein two), dimer named 22 - 1 - 1 antigen, the antigen is different from previously found in YH206, GA733, CA125, CEA tumor antigen, that is a new tumor associated antigen.Watanabe (Mol Cell Biol, 1998) to clone a new gene estrogen responsive in 1998 from human breast cancer cell line MCF-7 cDNA library, named EBAG9 (Estrogen receptor-binding fragment-associated gene 9), encoding region of 639bp, an estrogen response element in the upstream of 5', induce the expression of the mRNA molecule and protein with estrogen and upregulation of.Nakashima (Nat Med, 199 9) isolation and identification of genes encoding 22 - 1 - 1 antigens is named RCAS1 (receptor-binding cancer antigen expressed on SiSo cells). After sequence alignment, RCAS1 is actually the same protein as EBAG9.
Most research on the RCAS1 molecule is 22-1-1 monoclonal antibody by immunohistochemistry experiments in a variety of clinical tumor samples (such as cervical cancer, invasion of cervical squamous cell carcinoma, breast cancer, gastric cancer T3 expression, etc.) that RCAS1 expression and tumor differentiation, invasion and malignant degree of positive correlation, and prompt poor prognosis, there was no expression of RCAS1.RCAS1 can be used as the degree of malignancy in normal tissues
